摘 要:目的检测支气管哮喘患者外周血中白细胞介素32(Interleukin-32,IL-32)的表达水平,探讨其在支气管哮喘发病中的意义。方法收集35例支气管哮喘患者的外周血,收集25例健康人外周血作为对照组,用Trizol试剂盒提取外周血单个核细胞核糖核酸(RNA),用反转录试剂盒将RNA反转录为c DNA,实时荧光定量PCR(real-time PCR)检测IL-32 mRNA的表达;酶联免疫吸附试验(enzyme-linked immunosorbent assay,ELISA)检测IL-32蛋白质表达;统计学方法分析IL-32表达水平与免疫球蛋白E(Ig E)、嗜酸性粒细胞(eosinophile granulocyte,EOS)的相关性。结果支气管哮喘患者外周血中IL-32 mRNA及蛋白质表达均高于对照组(P<0.05)。Pearson相关性分析显示,哮喘患者外周血中IL-32表达水平与EOS、Ig E呈正相关(P<0.05),与降钙素原(procalcitonin,PCT)、C反应蛋白(C-reactionprotein,CRP)无相关性(P>0.05)。结论支气管哮喘患者外周血中IL-32表达增高,IL-32可能参与支气管哮喘的发病。Objective To investigate the level of serum interleukin- 32( IL- 32) and its significance peripheral blood in patients with asthma. Methods Thirty- five patients with asthma were enrolled in the present study. 25 healthy individuals were enrolled as control. Peripheral blood of patients and healthy individuals was collected. Total RNA of PBMCs was extracted using TRIzol. Then RNA was reversely transcribed into c DNA using reverse transcription Kit. Subsequently,real- time PCR was used to detect IL- 32 mRNA expression. Serum expression of IL- 32 protein was assayed by enzyme- linked immunosorbent assay( ELISA). Then the correlation of IL- 32 with Ig E and eosinophile granulocyte( EOS) was analyzed by using statistical method,respectively. Results IL- 32 RNA and protein expression levels in peripheral blood of asthma patients was higher than that in peripheral blood of the healthy control group( P〈0. 05). Pearson's correlation analysis showed that serum expression of IL- 32 in patients with asthma was positively correlated with EOS and Ig E,and was not correlated with procalcitonin( PCT) and C- reactionprotein( CRP)( P〉0. 05),respectively. Conclusion Serum IL- 32 expression increases in patients with asthma. So IL- 32 might be involved in the pathogenesis of asthma.
