通心降脂滴丸对犬冠脉结扎后心肌组织保护作用研究

摘　　要：目的观察通心降脂滴丸对犬冠脉结扎后冠脉血流量及心肌组织病理学改变的影响,探讨其对犬冠脉结扎后心肌组织的保护作用。方法取健康比格犬25只,♀♂兼用。按体重随机分为5组,每组5只,分别为假手术组、缺血模型组、阳性对照组(复方丹参片)、通心降脂滴丸低剂量组和高剂量组,于手术前1周开始给药,每日给药1次。高剂量组给予通心降脂滴丸0.503 g·kg-1,低剂量组给予通心降脂滴丸0.182g·kg-1,阳性对照组给予复方丹参片0.38 4 g·kg-1,假手术组和缺血模型组给予等容积的生理盐水;第8日进行手术,分离冠状动脉左前降支根部,放置Triton超声多普勒血流探头,测定冠状动脉血流量(CBF),术后将心脏取出,用10%的甲醛固定,HE染色法染色,分析实验结果。结果通心降脂滴丸低剂量组在给药后60、90、120 min时能增加冠脉血流量,与缺血模型组比较冠脉血流量变化百分率差异均有统计学意义(P<0.05)。通心降脂滴丸高剂量组在给药后30、60、90、120 min时能增加冠脉血流量,与缺血模型组比较冠脉血流量变化百分率差异均有统计学意义(P<0.05)。与缺血模型组相比,通心降脂滴丸高剂量组和阳性对照组心肌细胞损伤程度较低,主要表现为灶状水肿或轻度弥漫水肿,未发现炎细胞。结论通心降脂滴丸对犬冠脉结扎所致心肌缺血具有一定的保护作用。Objective To observe the impact of Tongxin Jiangzhi drops on the histopathological changes of myocardial tissues, and explore the protection of Tongxin Jiangzhi drops for myocytes in Beagle dogs with myocardial ischemia caused by coronary artery ligation. Methods Totally 25 male and female Beagle dogs were randomly divided into 5 groups： a sham operation group（0.9% Na Cl）, an ischemia model group（0.9% Na Cl）, a positive con trol group（0.384 g · kg- 1）, a Tongxin Jiangzhi drop low dose group（0.182 g · kg- 1）, and a high dose group（0.503 g · kg-1）. Medicines were given once daily for 1 we ek. The circumflex branch of the left coronary artery and root of aorta were separated. Ultrasonic doppler blood flow detector was put. When the surgery was finished, the myocardial tissue was collected for HE staining. Results Compared with the ischemia model group, the coronary blood flow at 60, 90 and 120 min increased in Tongxin Jiangzhi drops low dose group（P〈 0.05）. and the coronar y blood flow at 30, 60, 90 and 120 min increased in the Tongxin Jiangzhi drops high dose group（P〈 0.05）. Compared with the model group, the injury degree of myocardial cells in the Tongxin Jiangzhi drops high dose group and positive control group was lighter. The manifestations included belly like edema or mild diffuse edema. Edema inflammatory cells were not found. Conclusion Tongxin Jiangzhi drop has certain protective effect against myocardial ischemia induced by coronary artery ligation in Beagle dogs.
