摘 要:分离牛输卵管上皮细胞、培养、鉴定、核型分析和冷冻保存。第1代分离的上皮细胞(7±1)d时开始贴壁生长,(14.0±0.9)d后铺满培养瓶底部,第2代以后贴壁时间缩短为(1.0~1.5)d,体外培养至7代后逐渐衰老。第2代以后细胞贴壁性质发生了变化。解冻后细胞活率〉78%,输卵管上皮细胞呈角蛋白阳性,第5代细胞具有正常的二倍体核型,转染p EGFP-N3载体的效率较低。The epithelial cells of bovine oviduct were isolated,cultured and identified. The karyotype analysis was done and cells were cryopreservated. The results showed the first isolated epithelial cells began to adhere at( 7 ± 1) day,then covered the bottom at( 14. 0 ± 0. 9) day. From the 2nd generation the attaching time changed to 1. 0—1. 5 days. Till the 7th generation they began to senilite. After the 1st generation,the cellular adherent properties changed. The motility was above 78%.The oviduct epithelial cells expressed keratin positively. The 5th generation of epithelial cells contained normal diploid karyotype. The transfection rate of p EGFP-N3 was low.
