摘  要:目的:探讨解毒洗药对糖尿病大鼠创面肉芽组织中Smad3、Smad4表达的影响。方法:取50只Wistar大鼠制备糖尿病溃疡模型,将造模成功的18只随机分成3组,即解毒洗药组、贝复剂组、糖尿病模型组,每组6只;另取10只大鼠制备正常大鼠溃疡模型,选取造模成功的6只,设为正常模型组。各组分别给予相应处理后,应用免疫组化、Western Blot等技术观察各组大鼠创面肉芽组织中Smad3、Smad4表达水平的变化。结果:糖尿病模型组Smad3、Smad4表达水平明显高于解毒洗药组、贝复剂组和正常模型组(P<0.01);解毒洗药组Smad3、Smad4表达水平低于贝复济组(P<0.05),且与正常模型组比较,差异有统计学意义(P<0.05)。结论:Smad3、Smad4的高表达可能是糖尿病创面难以愈合的机制之一,解毒洗药可通过下调Smad3、Smad4的表达水平促进创面愈合。Objective: To investigate the effects of detoxicating wash on the expression of Smad3 and Smad4 in the wound granulation tissues of rats with diabetes. Methods: Fifty Wistar rats were treated to establish a model of diabetic ulcer. Eighteen successfully treated rats were randomly divided into three groups: detoxicating wash group,basic fibroblast growth factor( b FGF),and diabetes model group,with 6 rats in each group. Another 10 rats were treated to establish a rat model of ulcer,and the 6 successfully treated rats were set as normal model group. After respective treatments,immunohistochemistry and Western blot were used to measure the expression of Smad3 and Smad4 in wound granulation tissues. Results: The diabetes model group had significantly higher expression of Smad3 and Smad4 than the detoxicating wash group,b FGF group,and normal model group( P〈0. 01). The detoxicating wash group had significantly lower expression of Smad3 and Smad4 than the b FGF group( P〈0. 05),and there was a significant difference compared with the normal model group( P〈0. 05). Conclusion: High expression of Smad3 and Smad4 may be one of reasons that the wound seldom heals in diabetic patients. Detoxicating wash can promote wound healing by down- regulating the expression of Smad3 and Smad4.
