摘  要:目的通过观察大黄素、补骨脂素对辐射损伤模型小鼠的外周血有形成分、造血干/祖细胞和CD34+细胞表达的影响,探讨益髓生血颗粒治疗地中海贫血的药效物质基础。方法采用60Coγ射线3.5 Gy全身一次性照射,造成小鼠骨髓造血抑制、外周血象有形成分减少,致小鼠肾虚髓损。照射前大黄素、补骨脂素(60mg·kg-1,ig)预防给药2 d,造模后再连续给药14 d。在照射前及照射后的1,3,5,7,11,13,15,17,21 d检测外周血象;并在照后7d分别进行骨髓造血干/祖细胞集落培养(CFU-GM,CFU-E,BFU-E,CFU-Meg)及CD34+细胞检测。结果与照射对照组比较,从造模后第5天开始,G-CSF、大黄素、补骨脂素均表现出明显的促白细胞和血红蛋白升高的作用,但对红细胞均无明显的作用;在照射后7天,G-CSF、大黄素、补骨脂素均表现出显著促进CFU-GM,CFU-E的增殖的作用(P<0.001),而对CFU-GM,CFU-E均表现出轻微的抑制作用;此外,三组药物对照射后7 d的小鼠骨髓CD34+细胞均有促进增殖的作用,其中G-CSF(P<0.01)和大黄素(P<0.05)的作用具有统计学意义。结论大黄素、补骨脂素可一定程度的提高照射损伤小鼠外周血白细胞和血红蛋白的含量,可提高照射后小鼠骨髓造血干/祖细胞及骨髓CD34+细胞的增殖。研究结果提示,大黄素、补骨脂素是益髓生血颗粒的部分有效活性组分,具有提高小鼠造血功能的作用。Objective To explore the bioactive compounds of Thalassemia treated with Yisuishengxue granule by studying the effects of emodin,psoralen on express of visible components in peripheral blood,hematopoietic progenitor cells and CD34+cells in irradiated mice. Methods The60Coγ rays 3. 5Gy one-time illuminate the whole body,bone marrow hematopoietic inhibition and the reduction of peripheral blood elements in mice,cause the loss of kidney morrow. Emodin and psoralen( 60mg·kg-1,ig) was given as a prevention medicine before radiation for 2 days,and then successive administration for 14 days after successful modeling. Peripheral blood picture was examined before radiation and at the 1st,3rd,5th,7th,9th,11 th,13th,15 th,17th,19 th,21th days after radiation. Colonies of hematopoietic progenitor cells( CFU-GM,CFU-E,BFU-E,CFU-Meg,CFU-mix) were cultured and CD34+cell was examined at the 7th day after radiation. Results In the positive drug( G-CSF) group,emodin group and psoralen group,the count of the WBC and hemoglobin were increased from the 5th day after radiation compared with those in model group. But the three groups of drugs had no significant effect on red blood cells. At the 7th days after radiation,in the positive drug( G-CSF) group,emodin group and psoralen group,the proliferation of CFU-GM,CFU-E were significantly promoted,compared with model group(P〈0. 001),but the proliferation of CFU-GM and CFU-E were slightly inhibited. The positive drug( G-CSF) group(P〈0. 01) and emodin group(P〈0. 05) can obviously increased CD34+cell count in bone marrow of mice,compared with model group,and psoralen group also increased CD34+cell proliferation. Conclusion Emodin and psoralen can significantly increase the count of the WBC and hemoglobin,can promote proliferation of hematopoietic progenitor cells and increase CD34+cell count in bone marrow of mice.
