四种工艺提取的莪术油对Hela细胞增殖抑制的影响

摘　　要：目的探讨四种工艺从莪术中提取的挥发油对Hela细胞抑制作用,从疗效角度遴选最优工艺。方法分别采用石油醚浸提法、超临界CO2萃取法、压榨粗油经水蒸气蒸馏法精制、水蒸气蒸馏法提取莪术油,经标准品比对等纯度鉴定后,分别配制成浓度为10,20,40,80,160,320,640μg/ml的莪术油干预Hela细胞72h,MTT法检测细胞增殖抑制率及IC50的影响。结果四种工艺提取的莪术油对Hela细胞的生长均有明显抑制作用,最高抑制率分别为:石油醚浸提法莪术油为(87.48±4.33)%,超临界CO2萃取法莪术油为(57.56±4.75)%,鲜品压榨粗油经水蒸气蒸馏法精制莪术油为(94.13±5.36)%,水蒸气蒸馏法莪术油为(73.35±6.64)%,且抑制率呈现出一定的量效关系。其中压榨粗油经水蒸气蒸馏法精制莪术油在320μg/ml时对Hela细胞的抑制作用最强;IC50值依次为:131.79,253.62,98.79,223.82μg/ml。结论四种工艺提取的莪术油均可不同程度抑制Hela细胞的生长,压榨粗油经水蒸气蒸馏法精制莪术油对Hela细胞的抑制作用最强,提示压榨粗油经水蒸气蒸馏法精制可能为最佳工艺。Objective To study the inhibition effect to Hela cell of oil of zedoary turmeric,and filter out the optimum technology initially. Methods Respectively with the method of petroleum ether,supercritical CO2,extracted from pressed crude oil by distillation extraction method,water vapor to extract volatile oil from zedoary rhizome,purity identification with the standard substance,prepared the concentration to 10,20,40,80,160,320,640μg /ml,Hela cells were affected for 72 h,then cell proliferation inhibition rate and IC50 were observed by MTT assay,and the LDH content was detected. Results Oil of zedoary turmeric extracted by four methods significantly inhibited the growth of Hela cells. The highest inhibition rates were（ 87. 48 ± 4. 33） %,（ 57. 56 ±4. 75） %,（ 94. 13 ± 5. 36） %,（ 73. 35 ± 6. 64） % respectively,showing a good dose-dependent relation. The inhibiton of oil of zedoary turmeric by extracted from pressed crude oil by distillation extraction method is the most obvious at the concentration of320μg /ml; the IC50were131. 79 ug /ml,253. 62 ug /ml,98. 79 ug /ml,223. 82 ug /ml respectively. Conclusion The Hela cells could be differently inhibited by oil of zedoary turmeric extracted by steam,petroleum ether,supercritical CO2 and extracted from pressed crude oil by distillation extraction method,especially pressing method,maybe the pressing method is the optimum technology.
