复方丹参片对阿尔茨海默病转基因细胞模型AChE表达的影响

摘　　要：目的:研究复方丹参片对阿尔茨海默病(AD)转基因细胞模型乙酰胆碱酯酶(AChE)表达的影响。方法:将SD大鼠60只随机分成复方丹参片0.5、1、2、4倍剂量组,空白对照组及多奈哌齐对照组。各组大鼠分别灌服相应浓度药液,空白对照组给予等容积生理盐水,每天1次,连续7天。末次给药后1 h,取各组大鼠含药血清培养AD转基因细胞,于24、48、72 h采用Western Blot方法分析AD转基因细胞AChE的表达。结果:与空白对照组比较,24、48 h时2、4倍复方丹参片组AChE表达明显降低,差异有非常显著性意义(P<0.01),72h时差异无显著性意义(P>0.05);2、4倍复方丹参片组组内48h与24h比较,AChE表达降低,差异有非常显著性意义(P<0.01)。结论:复方丹参片能在一定程度上减少AD转基因细胞模型AChE的表达。Objective： To study the effect of compound Danshen tablets（CDT）on the expression of acetylcholinesterase （ACHE） in Alzheimer＇s Disease（AD） transgenic cell model. Methods： Sixty SD rats were randomly divided into 6 groups, namely 0.5- fotd, 1- fold, 2- fold, 4-fold CDT groups, blank control group and donepezil group. Rats in the medication groups were respectively given relevant concentrations of drugs, and the blank group was given same volume of normal saline, once a day, for 7 consecutive days. One hour after the last administration, serum containing medicine from each group was prepared to cultivate the AD transgenic cells. The levels of AChE protein were detected by Western blotting method after culturing for 24 h, 48 h, and 72 h. Results： Compared with the blank control group, the expression level of AChE was decreased in 2- fold and 4-fold CDT groups on culturing hour 24 and 48（P 〈 0.01）, while difference was insignificant on culturing hour 72（P 〉 0.05）, The intra-group difference of AChE level in 2- fold and 4-fold CDT groups was significant on culturing hour 24 and 48 respectively（P 〈 0.01）. Conclusion： Compound Danshen tablets can down-regulate the level of AChE in AD transgenic cell model to some degrees.
