摘 要:目的研究硫酸长春地辛对B淋巴细胞增殖及免疫功能的抑制作用。方法用CCK-8法检测不同浓度硫酸长春地辛(0、1、10、100、200、400ng/ml)作用于体外培养的多发性骨髓瘤XG-7细胞24 h后细胞活力的变化;同时用ELISA法检测细胞培养上清IgG水平及人B细胞活化因子(BAFF)水平;检测不同浓度硫酸长春地辛作用24 h后细胞的存活度。结果 (1)硫酸长春地辛作用24 h后,对XG-7细胞增殖有抑制作用,细胞活力随药物浓度增加逐渐降低,药物浓度为100、200、400 ng/ml时对细胞活力抑制作用明显,与空白对照组比较差异有统计学意义(P均<0.05);(2)当硫酸长春地辛浓度为100、200、400 ng/ml时细胞培养上清中IgG含量明显减少,与空白对照组相比差异有统计学意义(P均<0.05);(3)当硫酸长春地辛浓度为200、400 ng/ml时细胞培养上清中BAFF含量明显降低,与空白对照比较差异有统计学意义(P均<0.05);(4)不同浓度硫酸长春地辛对XG-7细胞的存活度均无明显影响(P均>0.05)。结论硫酸长春地辛能抑制XG-7细胞增殖,抑制其分泌IgG及BAFF,可能为免疫性疾病治疗提供一种新的途径。Objective To study the immunosuppressive effect of vindesine sulfate on B cells. Methods Human myeloma cell line XG-7 was cultured in vitro,and were treated with different concentration of vindesine sulfate( 1 ng /ml,10 ng /ml,100 ng /ml,200 ng /ml and 400 ng /ml) for 24 hours,The proliferation of XG-7 was detected by Cell Counting Kit 8( CCK8). The levels of IgG and B cell activating factor belonging to the TNF family( BAFF) in cell supernatants were measured by enzyme-linked Immunosorbent assay( ELISA). XG-7 cell viability was detected after treated with different concentration of vindesine sulfate. Results( 1) After vindesine sulfate effecting on XG-7 cell 24 h,the cell viability decreased gradually with the increase of vindesine sulfate concentration,and inhibition of cell viability was obvious at the vindesine sulfate concentrations of 100,200,400 ng /ml,there were significant differences compared with placebo group( all P〈0. 05).( 2) IgG levels in cell culture supernatants decreased at the vindesine sulfate concentrations of 100,200,400 ng /ml,there were significant differences compared with placebo group( all P〈0. 05).( 3) When vindesine sulfate concentrations was above 200 ng /ml,BAFF level in cell culture supernatant significantly decreased,there were significant differences compared with placebo group( all P〈0. 05).( 4) Different concentrations of vindesine sulfate showed no obvious effect on XG-7 cell survival. Conclusions Vindesine sulfate can inhibit XG-7 cell proliferation and IgG and BAFF secretion,which may provide a new approach for the treatment of autoimmune diseases.
