摘 要:目的通过探讨安宫牛黄丸对大鼠脑外伤后血脑屏障损伤及脑水肿的作用机制,旨在为临床研究安宫牛黄丸提高大鼠脑外伤及脑水肿的治疗效率提供理论依据。方法采用Marmarou方法制作大鼠弥漫性脑损伤模型,并给予安宫牛黄丸后应用HE染色、RT-PCR、免疫组织化学法及干湿质量法等方法统计脑组织含水量、脑组织梗塞面积、记忆提取能力、脑细胞凋亡及脑组织c-fos和c-jun的表达模式变化情况。结果应用安宫牛黄丸后大鼠脑组织含水量显著下降、脑组织梗塞面积减小、记忆提取能力提高、脑细胞凋亡数量减少,差异显著具有统计学意义(P<0.05)。脑组织c-fos和c-jun的表达模式呈现先增加后减少再增加的双峰模式。结论安宫牛黄丸治疗大鼠脑外伤后血脑屏障损伤及脑水肿,能显著减轻水肿、脑梗塞面积和脑细胞凋亡数,有效保护血脑屏障和脑组织。Objective To investigate the mechanism of Angong Niuhuang Pill on blood brain barrier injury and brain edema after traumatic brain injury of rats, in order to provide a theoretical basis for clinical study of Angong Niuhuang Pill improving the treatment efficiency of brain injury and brain edema of rats. Methods The rat model of diffuse brain injury was made by Marmarou, and the water content in brain, the infarction area of brain tissue, the memory retrieval ability of brain, the apoptosis of brain tissue, the situation changes of expression patterns of c-fos and c-jun of brain tissue were counted by HE-staining, RT-PCR, immunohistochemistry, dry wet weight methods after given the Angong Niuhuang Pill to rats. Results After the application of Angong Niuhuang Pill, the water content of rats brain tissue decreased significantly, the cerebral infarction area decreased, the memory retrieval ability increased, the number of the apoptosis of brain cells decreased, with statistically significant difference (P〈0.05). The expression pattern of c-fos and c-jun of brain tissue increased firstly, then decreased, and it increased again, appeared double peak model. Conclusions The treatment of Angong Niuhuang Pill on blood brain barrier injury and brain edema after traumatic brain injury of rats can significantly reduce the edema, cerebral infarction area and apoptosis of brain cells, and effectively protect the blood brain barrier and brain tissue.
