摘 要:目的研究茯苓多糖对异丙肾上腺素所致心肌肥厚大鼠心肌收缩功能和心肌舒张功能的影响。方法 Wistar雄性大鼠136只,体质量180-200 g。随机分为对照组(C-G,n=32)和造模组(M-G,n=104),其中C-G组随机分为C-G5周组(n=16)、C-G15周组(n=16);M-G组随机分为异丙肾上腺素诱导(I-G)5周组(n=11)、I-G 15周组(n=11);茯苓多糖高剂量(PCP-HG)5周组(n=14)、PCP-HG 15周组(n=14),茯苓多糖低剂量(PCP-LG)5周组(n=14)、PCP-LG15周组(n=14),卡托普利阳性对照(Cap-G)5周组(n=13)、Cap-G15周组(n=13)。给药方法为异丙肾上腺素皮下注射、卡托普利和茯苓多糖灌胃给予。采用水提醇沉法提取茯苓多糖,然后采用苯酚硫酸紫外分光光度法测定茯苓多糖。以异丙肾上腺素诱导建立大鼠心肌肥厚模型后给予茯苓多糖。观察各组大鼠心输出量(CO/BW)、左室收缩峰压(LVSP)、左室内压最大收缩率(+dp/dtmax)、左室舒张末压(LVEDP)、心脏指数(HMI)、左心室指数(LVMI)、心房利钠肽(ANP)和脑钠肽(BNP)的变化。结果I-G 5周组CO/BW200 mL·min^-1·kg^-1、ANP(260.08±0.19)pg/mL、BNP(466.14±25.75)pg/mL、HMI(4.64±0.45)g/kg、LVMI(3.14±0.25)g/kg,PCP-HG 5周组CO/BW 250 mL·min^-1·kg^-1、ANP(128.88±9.49)pg/mL、BNP(432.15±0.32)pg/mL、HMI(3.75±0.42)g/kg、LVMI(2.55±0.21)g/kg,两组比较差异均有统计学意义(P〈0.01)。I-G 15周组CO/BW 195 mL·min^-1·kg^-1、LVSP 80 mmHg(1 mmHg=0.133 kPa)、+dp/dtmax7 mmHg/s、LVEDP 18 mmHg、ANP(311.47±8.91)pg/mL、BNP(641.04±32.11)pg/mL、HMI(4.76±0.33)g/kg、LVMI(3.04±0.11)g/kg,PCPHG 15周组CO/BW 300 mL·min^-1·kg^-1、LVSP 130 mmHg、+dp/dtmax11 mmHg/s、LVEDP 7 mmHg、ANP(91.68±17.15)pg/mL、BNP(312.46±19.44)pg/mL、HMI(3.33±0.12)g/kg、LVMI(2.15±0.32)g/kg,两组比较差异均有统计学意义(P〈0.01)。PCP-HG5周组COObjective To explore effects of poria cocos mushroom polysaccharides( PCP) on myocaidial systolic and myocardial diastolic function of rats with isoproterenol-induced cardiac hypertrophy.Method One hundred and thirty-six Wistar rats( 180-200 g) were randomly divided into week 5 control group( C-G,n = 16),week 15 C-G( n = 16) and experimental group( M-G,n = 104). Experimental groups were randomly divided into Week 5( I-G,n = 11),Week 15 I-G,n = 11,week 5( PCP-HG,n =14) and week 15 PCP-HG,n = 14. PCP were extracted by ethanol subsiding method and the polysaccharides were measured by phenol-sulfuric acid visible spectrophotometry. The content of polysaccharides was 93.17%. Myocardial injury in mice was induced by subcutaneous injection of isoproterenol( I-G) and treated with intragastric PCP for 5,15 weeks( PCP-HG),hemodynamic parameters,CO /BW,LVMI,HMI,ANP and BNP were determined and analysed.
