摘  要:目的:探讨中草药在阿霉素肾病大鼠抗肾脏纤维化作用的实验研究。方法:雄性SD大鼠32只按随机数字表法分组。阿霉素肾病诱导模型方法分别使用贝那普利或加味当归补血汤(由黄芪、当归、川芎等组成,简称QGYS)在不同治疗组。监测血清白蛋白、血脂、24 h尿白蛋白和尿NAG酶的动态变化,以及肾脏体质量的比率测量和细胞外基质蛋白(ECM)在肾皮质的表达。实验结束时应用分子生物学方法分析转化生长因子β1(TGF-β1),基质金属蛋白酶组织抑制剂1(TIMP-1)和基质金属蛋白酶9(MMP-9)在肾皮质的表达。结果:QGYS与模型组大鼠比较可以提高血清白蛋白、降低血脂和肾的体质量比。同时减少尿蛋白排泄的水平和NAG酶以及调控细胞外基质(ECM)在肾皮质的集聚,QGYS同时调控TIMP-1、TGF-β1和MMP-9基因在肾脏表达水平。结论:QGYS能有效减少阿霉素肾病大鼠肾脏纤维化的程度,其作用效果可能是通过调节TIMP-1、MMP-9和TGFβ-1在肾脏的表达发挥作用。Objective: In order to investigate the anti-fibrotic effect of the Chinese herbs on adraimycin-induced nephropathy to delay the rate of renal fibrosis of rats. Methods : Thirty-two male rats of SD were randomly divided into four groups with 8 in each group. Nephropathy models were induced set up in the last latter three gourps induced by intravenously injection of adriamycin. Rats in the two treated groups were received intragastric administration with benazepri (a positive control) or Chinese herbs Qi Gui Yi Shen mixture (QGYS). Serum albumin, blood lipids, 24-h urine protein and urine N-acetyl-b-I)-glucosaminidase (NAG) were measured every two weeks. The ratio of kidney to body weight was measured too. The composition of renal tissue were analyzed by immunohistochemistry way, and transcription of genes encoding or molecular biology transforming growth factor ( TGF-[3~ ), the tissue inhibitor of matrix metalloproteinase 1 (TIMP-1) and matrix metalloproteinase 9 (MMP-9) was analyzed by RT-PCR at the end of the experiment. Results: Comparing to the untreated rats in the model group, QGYS can alleviate significantly serum albumin, decrease the blood lipids and the ratio of kidney to body weight. QGYS also significantly reduced the excretion levels of urinary protein and, N-acetyl-b-D-glucosaminidase (NAG) as well as and the accumulation of extracellular matrix (ECM) , in the renal tissue cortex. Additionally, QGYS decreased the gene expressions of TIMP-1 and TGF-bl, and increased MMP-9 gene expression. Conclusions: QGYS was effective drugs in treating in the rat model of nephropathy. The clinical benefit was associated with reduction of renal fibrosis. The anti-fibrotic effect of QGYS may be mediated through the regulation of TIMP-1, MMP and TGF-bl gene expressions. The anti-fibrosis function of QGYS may be correlated with the mechanism of regulate the ratio of enzyme which had the metabolism action of extracellular matrix (ECM) and cell factors.
