摘  要:目的:探讨蛇床子素对海人酸致痫大鼠海马神经元损伤的保护作用及相关分子机制。方法:将SD大鼠随机分为正常对照组、海人酸组和蛇床子素组,采用结晶紫染色法观察大鼠海马神经元的形态,采用免疫组化法检测大鼠海马神经元Puma蛋白的表达。结果:形态学观察结果显示,海人酸组较正常对照组大鼠的海马神经元排列不规则,数量减少,核固缩;蛇床子素组较海人酸组大鼠的海马神经元排列规则,数量增加,神经元形态与正常对照组相似。Puma蛋白检测观察结果显示,海人酸组较正常对照组大鼠海马神经元Puma蛋白表达增高(P<0.05),蛇床子素组较海人酸组大鼠海马神经元Puma蛋白表达降低(P<0.05)。结论:蛇床子素对海人酸致痫大鼠海马神经元损伤具有保护作用,其机制可能与下调大鼠海马神经元Puma蛋白表达有关。Objective: To investigate the neuroprotective effects of Osthole and related molecular mechanism against hippocampal neural damage in kainate- induced rats. Methods: SD Rats were divided into control group,kainate group and Osthole group. The morphology of the hippocampal neurons was observed with crystal violet staining. The expressions of Puma protein in hippocampal neurons was detected with immunohistochemistry staining. Results: The morphology observation showed that the hippocampal neurons in kainite group had unregular arrangement,number decreased compared with control group. The hippocampal neurons in Osthole group had regular arrangement,number increased compared with kainite group and the morphology in Osthole group was similar to that in normal control group. The detection of Puma protein showed that the expression of Puma protein in hippocampal neurons in kainate group was higher obviously to compare with control group( P〈0. 05). The expression of Puma protein in hippocampal neurons in Osthole group decreased obviously compared with kainate group( P〈0. 05). Conclusion: Osthole possesses the neuroprotective effects on kainate- damaged neuron,which may be related to downregulating the expression of Puma protein in hippocampal neurons.
