摘  要:目的探讨成骨诱导后的人脐血间充质干细胞(HUCBMSCs)移植对兔模型骨折愈合的影响。方法严格无菌条件下采集健康产妇志愿捐献的脐带血,采用密度梯度离心和贴壁培养法分离培养HUCBMSCs。取第3代细胞应用条件培养基进行成骨诱导2周;4-5月龄健康新西兰大白兔48只,体质量2.0-2.5 kg,雌雄不限,用线锯制作右胫骨横断骨折模型。将其分为A、B两组,每组又分为4个时点组,每小组6只。A组骨折断端注射成骨诱导后的HUCBMSCs;B组注射生理盐水。细胞移植后2、4、6、8周进行影像学、骨密度检测、组织学观察,观察评估骨折愈合情况。结果 2-8周X线片示:A组的骨折愈合速度要明显快于B组;组织学观察:A组新生骨小梁及类骨质明显多于B组;细胞移植后各时间点A组影像学结合组织学评分显著高于B组,两组比较差异有统计学意义(P〈0.05);骨密度检测:细胞移植后第2、4、6周A组骨密度高于B组,两组比较差异有统计学意义(P〈0.05),而在第8周A组骨密度低于B组,两组比较差异有统计学意义(P〈0.05)。结论成骨诱导后的HUCBMSCs移植能明显促进兔骨折愈合。Objective To discuss the role of the transplantation of human umbilical cord blood mesenchymal stem cells( HUCBMSCs) in rabbit fracture healing. Methods Records of 855 patients with primary cervical cancer from January 2002 to December 2012 were retrospective analysed. All patients underwent radiotherapy including external and cervical intracavitary irradiation: cassette four field with the dose of 30 Gy- 44 Gy was used in external irradiation,followed by the irradiation on parametrium with the total dose of 46Gy- 58 Gy,average dose was 36. 3 Gy in A point and 52. 7Gy in B point. Total 5- 6 times cervical intracavitary irradiation was performed with the dose 6 Gy- 7 Gy each time,average dose was 32. 6Gy in A point. 132 cases of them were treated with radical radiotherapy combined Iliac arterial interventional chemoembolization,Most of them received up to four cycles of chemotherapy. Results In 2- 8 weeks after surgery,X-ray film showed that the fracture healing of Group A was significantly faster than that of Group B. As for histological examination,the formation of new bone trabecular and sclerotin was significantly more in Group A than in Group B. After each time point,the scores of imaging combined with histological grading was significantly higher in Group A than in Group B( P〈0. 05). The bone mineral density of Group A was significantly higher than that of Group B( P〈0. 05). Conclusion The transplantation of HUCBMSCs after osteogenesis induction can significantly promote rabbit fracture healing.
