摘  要:【目的】观察大鼠后足切割后中枢神经系统内缩宫素(oxytocin,OT)及其受体(oxytocin receptor,OTR)的变化规律。【方法】雄性SD大鼠随机分为切割痛组(n=70)和对照组(n=20),切割痛组大鼠在切割后的30 min、1、3、6、24 h取脊髓腰膨大和下丘脑室旁核(paraventricular nucleus,PVN),检测OT、OTR阳性细胞表达,及OT、OTR转录和蛋白表达变化。【结果】与对照组大鼠相比,切割痛大鼠PVN内的OT阳性细胞数量及OT蛋白总量在切割后30 min^6 h明显下降,而m RNA却在切割后30min^1 h内升高;脊髓背角内OT阳性细胞数和蛋白表达量在切割后30 min^6 h上升,而OT m RNA无表达;切割前后PVN内均未见OTR的表达,而脊髓腰膨大背角Ⅰ、Ⅱ板层内OTR在切割后30 min^12 h表达上升。【结论】切割痛大鼠PVN内合成的OT可转运至脊髓发挥痛觉调制作用。【Objective】To investigate the variational regularity of oxytocin(OT) and itsoxytocin receptor(OTR) in the central nervous system ofin rats after hind paw incision.【Methods】Male SD rats were randomly divided into control group(group C, n=20) and incisional pain group(n=70). The number of OT and OTR-positve cells in paraventricular nucleus(PVN) and lumbar spinal cord was counted aton timepoint of 30 min、1 h、3 h、6 h and 24 h after incision. The expressions of m RNA and protein expression of OT and OTR were alsoin central nervous system was detected by RT-PCR, and the protein was detected by Western blotting.【Results】Compared to the group C, the number of OT-positive cells and expression of OT protein in the PVN of incisional rats with incisional pain decreased obviously from 30 min to 6 h after operation, however the m RNA increaseds from 30 min to 1 h;The reverse changes were detected in the spinal cord, where the OT-positive cells and protein expression increased from 30 min to 6 h without any m RNA transcription. The expression of OTR was negative in PNV but elevated in theⅠ-Ⅱlayers of lumbar spinal cord from 30 min to 12 h after incision.【Conclusions】The increased OT in the PVN of incisional rats with incisional pain may be transmitted to the spinal cord and play a modulation role of pain.
