HPLC法同时测定玉叶清火胶囊中栀子苷、穿心莲内酯和脱水穿心莲内酯

摘　　要：目的：建立玉叶清火胶囊中栀子苷、穿心莲内酯和脱水穿心莲内酯含量的液相色谱测定法。方法：采用Waters SunFire^TM C18色谱柱（4.6 mm×150 mm,5μm）,以甲醇-0.2%磷酸水溶液为流动相,梯度洗脱,流速1.0 mL·min^-1,检测波长为切换波长（0～10 min,238 nm,栀子苷;10～22 min,225 nm,穿心莲内酯;22～35 min,254 nm,脱水穿心莲内酯）。结果：栀子苷进样量在0.148～2.659μg范围内线性关系良好（r=0.999 9,n=6）,平均回收率（n=9）为99.90%;穿心莲内酯进样量在0.149～2.681μg范围内线性关系良好（r=0.999 8,n=6）,平均回收率（n=9）为99.62%;脱水穿心莲内酯进样量在0.089～1.607μg范围内线性关系良好（r=0.999 9,n=6）,平均回收率（n=9）为99.53%。结论：本文建立的波长切换高效液相色谱法简便、快捷,可用于该制剂的质量检测。Objective： To establish an HPLC method for the determination of geniposide,andrographolide and dehydroandrographolide in Yuye Qinghuo capsules. Methods： The Waters Sun Fire^TM C18（ 4. 6 mm × 150 mm,5 μm）column was adopted. The mobile phase was methanol-water containing 0. 2% phosphoric acid with gradient elution mode at the flow rate of 1. 0 mL·min^-1; the detection wavelength was 238 nm（ 0-10 min） for geniposide,225 nm（ 10-22 min） for andrographolide and 254 nm（ 22-35 min） for dehydroandrographolide. Results： Good linearity was shown in the range of 0. 148-2. 659 μg for geniposide with the average recovery（ n = 9） of 99. 90%,0. 149-2. 681 μg for andrographolide with the average recovery（ n = 9） of 99. 62%,0. 089-1. 607 μg for dehydroandrographolide with the average recovery（ n = 9） of 99. 53%. Conclusion： The wavelength switching-high performance liquid chromatography method is simple,rapid and convenient,which can be used for quality detection of Yuye Qinghuo capsules.
