‘黄花梨’2个CYP707A基因的克隆和序列分析

摘　　要：[目的]探讨脱落酸(Abscisic acid,简称ABA)分解途径关键酶基因(CYP707A)的特性,以期为揭示ABA调控梨花芽休眠进程的分子机制奠定基础。[方法]根据已公布的梨基因组数据库,通过本地blast的方法获得梨基因组CYP707A序列,运用DNAMAN软件设计引物,获得‘黄花梨’的2个脱落酸8’-羟化酶基因的CDS序列,分别命名为Pp CYP707A4(Genbank登录号:KP279630)和Pp CYP707A5(Genbank登录号:KP279631),并利用在线分析平台和生物软件进行生物信息学分析。[结果]Pp CYP707A4的CDS序列全长分别为1 431 bp,编码476个氨基酸;Pp CYP707A5的CDS序列全长为1 425 bp,编码474个氨基酸,两者同源性94.27%,氨基酸相似性94.96%,两者与苹果的2个基因在系统发育树上分别独自聚为一小支。[结论]Pp CYP707A4和Pp CYP707A5为同属于梨Pp CYP707A家族2个相似度较高的基因,它们可能发源于苹果和梨的共同祖先。[Objective] To understand the characteristics of CYP707 A that has been identified as the Abscisic acid 8＇-hydroxylase that was a key enzyme in ABA catabolic pathway,two CYP707 As were isolated from Pyrus pyrifolia‘Huanghua＇. [Method]The sequences of CYP707 A were retrieved from the dataset of pear genome project by homologues search using local blast,and the specific primers of CYP707 As were designed by DNAMAN. The genes were named as Pp CYP707A4（ Gen Bank Accession Number： KP279630） and Pp CYP707A5（ Genbank Accession Number： KP279631）. [Result]The main results of bioinformatics analysis were as follows： the full-length CDS of Pp CYP707A4 was1 431 bp which encoded a 476 amino acid polypeptide,and the open reading frame of Pp CYP707A5 was 1 425 bp which encoded a 474 amino acid polypeptide. Similarity comparisons showed that they share 94. 27% similarity in nucleotide sequence and 94. 96% similarity in amino acid sequence. The phylogenetic tree analysis showed that Pp CYP707A4 and Pp CYP707A5 were clustered lonely together with two CYP707 As from apple. [Conclusion]Pp CYP707A4 and Pp CYP707A5 may be similar and have originated in the common ancestor of apples and pears.
