摘  要:[目的]探讨菊花嵌合体‘su-07’花色变异的分子机理。[方法]以嵌合体植株的黄色花瓣和紫色花瓣为材料,采用凝胶电泳和生物质谱技术,对花色变异相关的差异表达蛋白质进行了分析。[结果]从不同花色花瓣的SDS-PAGE图谱中检测到了差异条带,在2-DE图谱中,检测到表达量差异在2倍以上的蛋白质斑点139个;选择其中表达量丰富且分离良好的16个蛋白质斑点进行质谱分析,成功鉴定了其中的10个蛋白质组分,分别涉及糖代谢、花色素代谢及基因调控等生命过程。[结论]嵌合花色的形成可能与这些蛋白质的差异表达有关。[Objective] The research aimed to investigate the molecular mechanism of color variation in chrysanthemum chimera ‘su-07 '.[Method]The differential expressed proteins related to color variation were analyzed through the gel electrophoresis and mass spectrometry by using chimera petals( yellow and purple petals) as experimental materials. [Result] The differential bands were detected from the different color petals based on the SDS-PAGE images. And in two-dimensional electrophoresis( 2-DE) images,the expressed levels of 139 protein spots differed significantly. Sixteen spots with high expression levels and well separation were analyzed by mass spectrometry,and ten of them were identified successfully. The proteins are involved in the glycometabolism,anthocyanin metabolism and gene regulation and other life processes.[Conclusion]The formation of chimera may be related to differential expression of these proteins.
