摘  要:以5、7、9、11月取材的地涌金莲幼嫩花蕾为外植体,对不同取材时期愈伤组织诱导率、褐化率及分化率进行研究。结果表明,5月取材的外植体愈伤组织诱导率最低,为67.79%,显著低于其余取材时期,11月取材的外植体愈伤组织诱导率最高,达83.96%,7月与9月取材的相互间无显著差异,但均显著高于5月取材的而显著低于11月取材的;从5月至11月取材的外植体愈伤组织褐化率有逐渐降低趋势,5月的最高,达78%,与7月取材的无显著差异,但显著高于9月、11月的63.5%和60%;9月、11月取材的外植体愈伤组织分化率最高,达到17.47%和20.12%,相互间无显著差异,但均显著高于5月和7月的10.67%和13.11%。因此,在实际生产中以秋季取地涌金莲幼嫩花蕾为外植体进行组织培养较为适宜。Callus induction rate,browning rate and differentiation rate of different sampled time in May,July,September and November to the tender buds of Musella lasiocarpa explants were studied. The results showed that the lowest rate of callus induction was sampled in May,and it was 67. 79 %,significantly lower than the others; the highest was 83. 96 % in November; there were no significant differences between July's and September's,but they were both significantly higher than May's and significantly lower than November's. As to callus browning rate,it had gradually decreased from May to November; the highest browning rate was sampled in May,up to 78 %; though was not obviously different with July's,significantly higher than September's( 63. 5 %) and November's( 60 %). As regard to callus differentiation rate,the highest one was sampled in November,reaching 20. 12 %,and it has no significant difference with September's( 17. 47%),but they were significantly higher than May's( 10. 67 %) and July's( 13. 11 %). This research showed that the tender buds of Musella lasiocarpa should be sampled in autumn when they were used as explants for tissue cluture.
