摘  要:目的探讨日本血吸虫(Schistosoma japonicum,SJ)感染鼠树突状细胞(Dendritic cell,DC)亚型对过敏性哮喘的抑制作用及其机制。方法采用MACS方法从SJ感染鼠脾脏中提取CD8α+DC(SJCD8α+DC)和CD8α-DC(SJCD8α-DC),分别过继转移给BALB/c小鼠,并用OVA诱发哮喘(SJCD8α+组和SJCD8α-组),以正常组(Normal组)和单纯诱发哮喘组(Asthma组)作为对照。取各组小鼠肺组织作石蜡切片,HE染色后观察肺组织病理变化。采用qRTPCR检测SJCD8α+DC和SJCD8α-DC中IL-12和IL-10mRNA水平,以正常鼠CD8α+DC(NCD8α+DC)和正常鼠CD8α-DC(NCD8α-DC)作为对照。用ELISA方法检测各组小鼠支气管肺泡灌洗液(Bronchial alveolar lavage fluid,BALF)和脾细胞培养上清中IL-10水平。结果肺组织病理学检查显示DC过继转移组过敏性哮喘被抑制,其中SJCD8α-组的抑制效果强于SJCD8α+组。SJCD8α-DC IL-10mRNA表达水平高于SJCD8α+DC(P<0.05)。并且,SJCD8α-组的BALF和脾细胞培养上清中IL-10蛋白表达水平明显高于SJCD8α+组(P<0.01,P<0.01)。SJCD8α-DC和SJCD8α+DC的IL-10mRNA水平分别为1.73±0.24和1.03±0.03(GAPDH%)。SJCD8α-组和SJCD8α+组的BALF IL-10蛋白水平分别为411.21±6.38pg/ml和202.88±25.30pg/ml,两组的脾细胞培养上清中IL-10蛋白水平分别为841.53±7.75pg/ml和254.16±21.46pg/ml。结论过继转移SJCD8α-DC可能通过高表达IL-10抑制OVA诱导的过敏性哮喘。Objective To explore the inhibitory effect of dendritic cell(DC)subsets from mice infected with Schistoso majaponicumon allergic asthma and their mechanism of action. Methods CD8α+DC and CD8α-DC were extracted from spleens of mice infected with S.japonicumusing MACS(SJCD8α+DC and SJCD8α-DC)and were adoptively transferred to BALB/c mice(an SJCD8α+group and an SJCD8α-group).Then,allergic asthma was induced with OVA.Normal mice(the Normal group)and untreated mice with induced asthma(Asthma group)served as controls.The lungs of the different groups of mice were histologically analyzed using HE staining.Levels of IL-12 and IL-10 mRNA were measured in CD8α+DCs and CD8α-DCs using qRT-PCR.Levels of IL-10 protein in bronchial alveolar lavage fluid(BALF)and the spleen cell culture supernatant from different groups of mice were measured using ELISA. Results OVA-induced allergic asthma was inhibited in groups receiving DCs from mice with schistosomiasis.The SJCD8α-group had a markedly inhibited lung inflammatory response compared to the SJCD8α+group.The level of IL-10 mRNA was higher in SJCD8α-DCs than in SJCD8α+DCs(P〈0.05).In addition,the level of IL-10 protein in both BALF and the spleen cell culture supernatant was higher in the SJCD8α-group than in the SJCD8α+group(P〈0.001,P〈0.001).The level of IL-10 mRNA was 1.73±0.24 in SJCD8α-DCs and 1.03±0.03 in SJCD8α+DCs(expression/GAPDH%).The level of IL-10 protein in BALF was 411.21±6.38pg/ml in the SJCD8α-group and 202.88±25.30pg/ml in the SJCD8α+group.The level of IL-10 protein in the spleen cell culture supernatant was 841.53±7.75pg/ml and 254.16±21.46pg/ml for the two groups. Conclusion Adoptive transfer of CD8α-DCs from mice infected with S.japonicuminhibited OVA-induced allergic asthma by increasing the production of IL-10.
