摘　　要：目的研究马钱苷对结肠癌SW480细胞增殖的影响及作用机制。方法 MTT法检测细胞增殖和生存活力;Hoechst 33258染色后荧光显微镜观察细胞形态,流式细胞仪检测细胞凋亡;半定量RT-PCR法检测与细胞凋亡相关的Bax基因和Bcl-2基因mRNA的表达水平。结果与对照组比较,不同浓度药物组均对SW480细胞有抑制作用,且呈一定的量效和时效关系;Hoechst 33258染色可见较多凋亡细胞,流式细胞仪检测显示马钱苷能显著增加细胞凋亡率;半定量RT-PCR测得马钱苷对Bax mRNA的表达无影响,但会引起Bcl-2 mRNA的表达下调。结论马钱苷能抑制SW480细胞的增殖,并诱导细胞凋亡,其作用机制可能与影响Bcl-2基因的表达有关。Objective To investigate the effect and mechanism of Loganin on the proliferation of colon cancer SW480 cells. Methods Cell proliferation and viability were assessed by MTT assay. Hoechst 33258 staining was performed to observe the morphologic changes of cells. The cell apoptosis rate was tested by FCM.Real-time PCR was used to detect the expression of Bax and Bcl-2 mRNAs. Results Compared with the untreated group,Loganin treatment inhibited the proliferation of SW480 cells in a time-and dose-dependent manner. Hoechst 33258 staining displayed typical morphological changes. Loganin significantly increased the apoptosis rate of SW480 cells by flow cytometry. Meantime,Loganin reduced Bcl-2,but not Bax,mRNA expression at 24 h in SW480 cells. Conclusion Loganin can inhibit the proliferation and induce apoptosis on SW480 cells,which may be related to reduction of Bcl-2 mRNA expression.
