摘  要:目的:基于i TRAQ技术探讨高脂血症大鼠肝脏蛋白质组学特征,为进一步从分子水平探讨高脂血症大鼠的病理机制提供新的理论依据。方法:20只健康雄性SD大鼠随机分为正常组和高脂血症组,正常组每日给予普通大鼠维持饲料;高脂血症组给予高脂饲料饲喂,30 d后,检测大鼠血脂情况和肝脏组织形态学变化,进行模型评估。分离提取肝脏总蛋白,经过蛋白质酶解、i TRAQ标记后,先后采用岛津LC-20AB液相系统进行液相分离和岛津公司LC-20AD型号的纳升级液相色谱仪进行分离。然后采用串联ESI质谱仪对经过液相分离的肽段进行分析,最后,用Mascot 2.3.02软件在蛋白质数据库中搜索肽质量指纹谱数据进行肽段及蛋白质的鉴定并确定蛋白质。结果:统计分析结果显示,与正常组相比,高脂血症大鼠肝脏中173个蛋白上调,312个蛋白下调,本文中我们挑选出4个涉及脂类代谢的蛋白加以分析。结论:高脂血症的发生可能与单酰甘油脂肪酶、低密度脂蛋白受体相关蛋白上调,载脂蛋白A-I、羧酸酯酶3下调有关。Objective: To observe proteomic features in the liver of hyperlipidemia rats by i TRAQ technology,provided a new theoretical reference for the further study on the molecular level to explore the pathological mechanism of hyperlipidemia rats. Methods: Twenty healthy male SD rats were randomly assigned into normal group and hyperlipidemia group.Hyperlipidemia group was fed with high- fat diet,while blank control group was fed with normal diet. 30 days later,Levels of serum cholesterol( TC),triglyceride( TG),low density lipoprotein cholesterol( LDL- C) and high density lipoprotein cholesterol( HDL- C) were detected by fully automatic biochemical analyser. HE and oil red staining methods were used to observe the liver injury. Extraction of liver total protein,after protein enzymolysis,labeled by i TRAQ,SHIMADZU LC- 20 AB liquid phase system and LC- 20 AD Nano liquid chromatography instrument were used to separate the components. Finally,Identified peptide and protein by searching peptide mass fingerprinting data in protein database and determined the protein by Mascot 2. 3. 02 soft. Results: The statistical analysis found that compared with normal group,173 proteins were up- regulated and 312 proteins were down- regulated in Hyperlipidemia group,and we selected 4 proteins involved in lipid metabolism to analysis. Conclusion: The occurrence of hyperlipidemia may be associated with the up- regulation of LRP and MAGL、the down- regulation of apolipoprotein A- I and carboxylesterase 3.
