摘  要:目的:选择一种准确、简便的测定瓜蒌皮精氨酸含量的方法。方法:对异硫氰酸苯酯(PITC)和邻苯二甲醛(OPA)两种柱前衍生方法测定精氨酸的含量进行比较。结果:PITC衍生法有副反应产物,干扰测定结果。OPA衍生法简化了衍生过程,衍生方便、快速,衍生产物单一,测定结果相对准确。结论:PITC衍生法测定瓜蒌皮中精氨酸含量不准确,操作繁琐,不可作为测定瓜蒌皮精氨酸含量测定的方法。OPA衍生法测定瓜蒌皮中精氨酸含量较准确,操作简便,可为中药精氨酸含量测定方法选择提供参考。Objective: To choose an accurate and simple method for the determination of arginine in Trichosanthis Pericarpium. Methods: The phenylisothiocyanate( PITC) and o- Phthalaldehyde( OPA) pre-column derivatization methods were compared by determining the content of arginine. Results: PITC derivative method had side products,which interfered the determination results. OPA method which simplified the derivation process was convenient,fast and single and the results were relatively accurate. Conclusion: PITC derivatization had vice reaction products and was not accurate for the determination of arginine in Trichosanthis Pericarpium and can not be used for the determination of arginine in Trichosanthis Pericarpium. OPA derivatization was relatively accurate and simple and can provide the reference for arginine determination in traditional Chinese medicine.
