摘 要:目的:通过对哮喘小鼠肺组织水通道蛋白(AQP)的影响,探讨AQP与中医“痰”的联系及麻杏二三汤抑制哮喘小鼠气道黏液高分泌的机制。方法:雌性BALB/c小鼠60只,随机分成:1空白对照组,2哮喘模型组,3强的松组,4中药(麻杏二三汤)高剂量组,5中药中剂量组,6中药低剂量组。以腹腔注射0.1%卵白蛋白(OVA)与10%氢氧化铝混悬溶液致敏,以1%OVA雾化液雾化攻击制作哮喘小鼠模型。1-6组分别给予生理盐水、生理盐水、强的松(10.0 mg/kg)、麻杏二三汤高剂量(30 g/kg)、中剂量(15 g/kg)、低剂量(7.5 g/kg)灌胃,免疫组化法测定各组AQP1、AQP5在肺组织中的分布和表达。结果:麻杏二三汤高、中、低剂量组AQP1、AQP5表达弱于模型组(P〈0.01或P〈0.05)。结论:麻杏二三汤能够有效抑制哮喘小鼠气道黏液高分泌的机制可能与其能够调节支气管肺组织中AQP5、AQP1的表达有关。Objective: Through experiments to study the effect of Maxing Ersan Decoction on the asthmatic mice's lung aquaporins( AQP) and to discuss the relationship between the AQP and the phlegm of traditional Chinese medicine and the mechanism of Maxing Ersan Decoction inhibiting the asthmatic mice'airway mucus hypersecretion. Methods: Taking60 female BALB / c mice with weight between 18 - 22 g were randomly divide into 6 groups: the blank control group,the asthma model group,the dexamethasone group,the Maxing Ersan Decoction high dose group,the Maxing Ersan Decoction medium dose group,the Maxing Ersan Decoction low dose group. Intraperitoneal injection by the fresh mixture of 0. 1%ovalbumin( OVA) and 10% Aluminum hydroxide suspension solution was used to make the mice sensitization and atomizing with 1% OVA atomized liquid to induce asthma. The first to the sixth group were respectively gavaged by NS,NS,prednisone( 10. 0 mg / kg),Maxing Ersan Decoction( 30 g / kg),Maxing Ersan Decoction( 15 g / kg),Maxing Ersan Decoction( 7. 5 g / kg). The immunohistochemical method was used to detecte the AQP1 and AQP5's distribution and expression in lung tissue of each group. Results: The expressions of the AQP1 and AQP5 were positive in the asthma model group and stronger than those of the blank control group with similar parts. Each of the Maxing Ersan Decoction groups was weaker than that of the asthma model group( P〈 0. 01 or P〈 0. 05). Conclusion: The mechanism of Maxing Ersan Decoction can effectively inhibit the asthmatic mice's airway mucus hypersecretion,which may be related to regulating the expressions of AQP5 and AQP1.
