姜黄素抑制单侧输尿管梗阻大鼠肾间质纤维化的实验研究

摘　　要：目的:观察姜黄素(Curcumin)对单侧输尿管梗阻(UUO)模型大鼠肾间质纤维化的拮抗作用。方法:30只雄性大鼠随机分为假手术组、UUO模型组和姜黄素组,每组10只,术后第6天检测24 h尿蛋白定量,于术后7 d处死大鼠,留取梗阻侧肾组织,检测血清肌酐(Scr)、尿素氮(BUN)、胱胺酸蛋白酶抑制剂C(Cystain C)、行Masson染色观察肾间质纤维化程度,免疫组化染色检测肾间质Ⅰ型胶原(ColⅠ)、α-平滑肌肌动蛋白(α-SMA)的表达。结果:与模型组相比较,姜黄素组Scr、BUN、Cystain C水平显著降低(P<0.05),肾间质纤维化相对面积显著减少(P<0.05),α-SMA及ColⅠ的表达明显降低(P<0.05)。结论:姜黄素可以减少输尿管梗阻后α-SMA及ColⅠ的表达,从而改善UUO所致的肾脏损伤,发挥其抗肾间质纤维化作用。Objective： To observe the protective effects of Curcumin on renal interstitial fibrosis of rat model with unilateral ureteral obstruction（ UUO）. Methods： Thirty male SD rats were randomly divided into 3 groups： sham-operation group,the UUO model group and Curcumin treatment group,10 rats for each group. All rats were sacrificed at the seventh day after the operation. The level of 24 hours＇ urinary protein excretion was determined one day before the rats were sacrificed. The obstructed kidney was taken for histology evaluation and molecular biology assay,Scr,BUN and Cystain C were determined for each group. Masson stain was performed as a main indicator of interstitial fibrosis. The expression of α-smooth muscle actin（ a-SMA）,and collagen I in renal tissue was detected by immunohistochemistry staining,respectively. Results： Compared with those of model group,levels of Scr,BUN,Cystain C were significantly decreased in Curcumin treatment group（ P 〈0. 05）,and the area of interstitial fibrosis was significantly decreased（ P 0. 05）,the expression of collagen I and α-SMA was significantly down-regulated（ P 〈0. 05）. Conclusions： Curcumin could decrease the interstitial fibrosis by inhibiting epithelial to mesenchymal transition in UUO rats.
