扶正活血中药治疗放射性肺损伤的实验研究

摘　　要：目的观察放射性肺损伤大鼠肺组织转化生长因子-β1（TGF—β1）表达水平及肺组织病理学特征，探讨扶正活血中药对放射性肺损伤的防治作用及可能机制。方法将56只SD雌性大鼠随机分为模型组、前期干预组、后期短程干预组及后期长程干预组，每组14只，通过单次大剂量6MV—X线加速器胸部照射制成放射性肺损伤大鼠模型。分别不用及照射前2周内、照射后1个月内、照射后3个月内用扶正活血中药灌胃，于照射后3、6个月分别随机处死各组大鼠7只，取大鼠肺组织进行免疫组化检测TGF-β1蛋白表达强度及观察不同时相肺组织病理学特征。结果各干预组大鼠肺组织TGF—β1蛋白表达强度明显低于模型组，放射性肺损伤病理学改变明显轻于模型组。结论扶正活血中药具有抑制TGF—β1分泌，有效阻止或延缓肺纤维化的形成，对放射性肺损伤的防治具有一定作用。Objective To investigate the effect of Chinese medicine of Fuzheng Huoxue on radiation-induced lung injury in rats. Methods Fifty six S-D rats were randomly allocated into irradiation group（n=14）, early intervention group（n=14）, short-term intervention group （n=14）, long-term intervention group （n=14）. The radiation lung injury was induced by irradiation on chest of rats with a single large dose using 6-MV linear accelerator. Chinese medicine of Fuzheng Huoxue was administrated by gavage at 2 weeks before irradiation（early intervention group）, within 1 month or 3 months after irradiation（short-term or long-term intervention groups）, respectively. The animals were sacrificed at the end of the 3 and 6 months after irradiation （7 in each batch）. The histopathological changes of lungs was observed and the expression of TGF- β1 in lung tissue was detected with immunohistochemistry. Results The expression of TGF-β1 in intervention groups was significantly lower than that in model group. Compared to model group the histopathological changes of radiation-induced lung injury were significantly ameliorated in intervention groups. Conclusion Chinese medicine of Fuzheng Huoxue can alleviate or delay the radiation-induced lung injury in rats, which may be associated with the down-regulation of TGF-β1 ; expression in lung tissue.
