摘 要:目的:观察解表药桂枝不同萃取部位的抗过敏作用,探讨该药抗过敏作用的有效物质基础,为其功效和临床应用提供药理依据。方法:采用2,4-二硝基氯苯(DNCB)诱发的小鼠迟发型超敏反应模型,及小鼠异种被动皮肤过敏反应模型观察各受试药液对模型动物毛细血管通透性的影响,以初步筛选有效部位。进一步采用DNCB致小鼠迟发型超敏反应以及组胺、5-羟色胺所致小鼠皮肤毛细血管通透性增高模型,和体外透明质酸酶抑制实验验证桂枝去油水煎液、乙酸乙酯部位与水提部位的抗过敏作用。结果:桂枝去油水煎液(6g生药/kg)、乙酸乙酯萃取部位(6g生药/kg)、水提部位(6g生药/kg)能显著降低迟发型过敏反应模型小鼠的毛细血管通透性,乙酸乙酯部位组亦能明显抑制该模型小鼠的耳廓肿胀度;三者对小鼠异种被动皮肤过敏反应有明显抑制作用。桂枝去油水煎液及乙酸乙酯部位对组胺、5-HT所致小鼠毛细血管通透性的增加有显著抑制作用;两者在浓度0.5g/ml时,体外对透明质酸酶有较强的抑制作用,抑制率分别为52.48%、52.17%。结论:桂枝去油水煎液、乙酸乙酯部位具有较好的抗过敏作用,提示桂枝抗过敏作用的有效组分以极性较高的水溶部位为主,其作用机制与抗过敏介质(如His、5-HT)效应有关。Objective: To observe the antiallergic effect of nourish of different extractions of Cinnamomi Ramulus, so as to explore the effective ma- terial base of its antiallergic effect and provide pharmacology basis for its efficacy and clinical application. Methods: By using the model of delayed hypersensitivity induced by DNCB in mice, and the model of passive cutaneous anaphylaxis reaction in mice, to preliminarily screen the effective extraction through the index of capillary permeability. Further by using the model of late hypersensitivity induced by DNCB in mice, and the model of skin capillary permeability increasing induced by histamine or 5- serotonin in mice , and the experiment of hyaluroni- dasee inhibition in vitro, to validate the antiallergy effect of the above effective extractions. Results: The water decoction of Cinnamomi Ram- ulus without oil (6g crude drug/kg), the extraction of ethyl acetatefractions (6g crude drug/kg) and the extraction of water (6g crude drug /kg) significantly reduced the capillary permeability in delayed hypersensitivity reaction model in mice (P 〈0.01 or P 〈 0.05 ), as the ex- traction of ethyl acetatefractions significantly reduced the ear swelling degree in delayed hypersensitivity reaction model in mice ( P 〈 0.05), and the above three groups also significantly inhibit the skin capillary permeability of passive cutaneous anaphylaxis reaction in mice ( P 〈 0. 01 or P 〈 0.05). The water decoction of Cinnamomi Ramulus without oil and the extraction of ethyl acetatefractions significantly reduced the increasing of skin capillary permeability induced by histamine and 5-HT in mice ( P 〈 0.01, P 〈 0.05 ), and the two extractions all showed high inhibition rate on hyaluronidase under the concentration of O. 5g/ml in vitro, which the inhibition rate of the water decoction of Cinnamo- mi Ramulus without oil and the extraction of ethyl acetatefractions were 52.48% and 52.17% respectively.
