黄芪提取物诱导人胃癌SGC-7901细胞凋亡的机制研究

摘　　要：目的:研究黄芪提取物对人胃癌SGC-7901细胞凋亡的影响及其作用机制。方法:采用不同浓度的黄芪提取物干预人胃癌SGC-7901细胞。MTT比色法观察黄芪提取物对细胞增殖的影响;流式细胞仪检测细胞凋亡率和线粒体膜电位;分光光度法检测细胞Caspase-3和Caspase-9活性;电泳法检测Bax和Bcl-2蛋白表达。结果:黄芪提取物(25、50、100 mg/L)以剂量依赖性地抑制SGC-7901细胞增殖,诱导其凋亡,并且降低线粒体膜电位;增加Caspase-3和Caspase-9活性,上调Bax蛋白表达的同时下调Bcl-2蛋白表达。结论:黄芪提取物可抑制人胃癌SGC-7901细胞体外增殖且通过线粒体途径诱导其凋亡。Objective： To investigate the effects and mechanism of Astragalus extract on apoptosis of human gastric cancer SGC-7901 cell in vitro. Methods ： The effects of different concentrations of Astragalus extract on human gastric cancer SGC-7901 cell were observed. Proliferation in- hibiting rate was measured by MTr assay; The apoptosis and mitochondrial membrane potential were analyzed by flow cytometer. The activi- ties of Caspase-3 and Caspase-9 were detected by spectrophotometry. The protein expressions of Bax and Bcl- 2 were studied by western blot- ting. Results： Astragalus extract（25,50,100mg/L） dose-dependently inhibited the proliferation of human gastric cancer SGC-7901 cell, in- duced its apoptosis and decreased the mitochondrial membrane potential, enhanced the activities of Caspase-3 and Caspase-9, increased the Bax protein expression while decreased the bcl-2 protein expression. Conelusions：Astragalus extract can inhibit the proliferation of human gastric cancer SGC-7901 cell and induce its apoptosis through mitochondrial pathway in vitro.
