摘  要:目的:筛选青果中抗炎镇痛的活性部位,探讨活性部位对细胞炎性因子IL-1β、IL-10、TNF-α蛋白表达的影响。方法:分别用水蒸气蒸馏法、溶剂梯度萃取法获得青果挥发油、青果10%水提液及其石油醚、氯仿、乙酸乙酯、正丁醇萃取部位。首先采用二甲苯致小鼠耳廓肿胀、角叉菜胶致大鼠足跖肿、醋酸致扭体反应和小鼠热板性疼痛筛选抗炎镇痛的活性部位,然后采用Western blot检测活性部位对炎性因子IL-1β、IL-10、TNF-α蛋白表达的影响。结果:青果水提液及其石油醚、氯仿、乙酸乙酯萃取部位800mg/kg均能减轻二甲苯致小鼠耳廓肿胀和角叉菜胶致大鼠足跖肿胀,提高小鼠对热板性疼痛的痛阈值,减少扭体反应的平均次数,其中乙酸乙酯萃取部位活性最强,但青果挥发油和正丁醇萃取部位800 mg/kg无明显抗炎镇痛作用。青果水提液的乙酸乙酯萃取部位400 mg/kg、800 mg/kg、1200 mg/kg显著降低角叉菜胶致足跖肿胀模型大鼠细胞因子IL-1β和TNF-α的蛋白表达,但对IL-10表达无明显的影响。结论:青果水提液的乙酸乙酯萃取部位是抗炎镇痛的活性部位,其作用机制可能与抑制细胞炎性因子IL-1β和TNF-α的表达有关。Objective: To Screen the anti-inflammatory and analgesic active site from Fructus Canarii and Observe the effect of active site on Inflam- matory cell factor IL-1β,IL-10 and TNF-a. Methods:The essential oil of Fructus Canarii was extracted by steam-stilling. Other solvent ex- tracts were obtained by extracting Fructus Canarii'water - soluble part with petroleum ether, chloroform, ethyl acetate and n-butanol in order.Using mouse auris-swell model resulted in dimethylbenzene, the model of rat paw edema induced with carrageenan, writhing induced with acetic acid in mice and Hot plant test as screening to anti-inflammatory analgesic active site, further using Western blot analyses to detect ac- tive site of inflammatory cell factor IL-1β, IL-10, TNF-ct. Results: Fructus Canarii'water - soluble part with ether, chloroform, ethyl acetate extract 800 mg/kg could reduce mouse auricle swelling induced by xylene and pelvetia silquosa glue to rats foot metatarsus swelling, increase the mice pain threshold of hot plate pain, reduce the average number of twisting body response, including petroleum ether extraction site most, but the essential oil of Fructus Canarii and n-butanol extraction parts 800 mg/kg have no obvious anti-inflammatory analgesic action. Petrole- um ether extraction parts 400 mg/kg,800 mg/kg and 1200 mg/kg significantly down-regulated the expression of cytokines IL - 1 and TNF -a in the model of rat paw edema induced with carrageenan, but had no obvious effect on the expression of IL - 10. Conclusions: Fructus Cana- rii' water - soluble part with petroleum ether is anti-inflammatory analgesic active site, its mechanism may be related to inhibiting the expres- sion of inflammatory cell factor IL - 1βand TNF-a.
