犬椎间盘内注射医用三氧对NF-κB表达水平的影响

摘　　要：目的 观察犬椎间盘内注射医用三氧对NF-κB表达的影响.方法 选择健康本地犬15只,随机分为3组,每组5只.在DSA引导下行L1～2～L4～5椎间盘穿刺.A组（对照组）仅穿刺,不注射任何物质;B组（纯氧组）注射纯氧5 ml;C组（医用三氧组）注射30～50 μg/ml医用三氧5 ml.所有动物于操作后1周处死,取椎间盘组织,分别采用RT-PCR和Western-blot方法测定其中NF-κBp65mRNA与NF-κBp65蛋白的表达.结果 RT-PCR结果显示医用三氧组髓核细胞内NF-κBp65mRNA及蛋白表达水平均低于对照组与纯氧组,差异有统计学意义（P＜0.05）.结论 犬椎间盘内注射医用三氧可降低椎间盘髓核细胞内核转录因子NF-κB的表达.Objective To investigate the effects of intervertebral disc injection with medical ozone on the expression of NF-nB in canine. Methods Fifteen healthy adult canines were randomly divided into three groups （n=5） ： the control group（group A）, the pure oxygen group （group B） and the medical ozone group （group C）. In group A the canines were punctured percutaneously to the in- tervertebral disc under DSA without any medication injected; pure oxygen was injected into interver- tebral discs in the group B; while 30-50 μg/ml ozone was injected into intervertebral disc in the group C. All the canines were sacrificed at 1 week after the management. The lumbar discs were col- lected to detect the protein expression of NF-kBp65 by Western blot and NF-kBp65mRNA by RT- PCR. Results The expressions of NF-kBp65mRNA and protein were significantly lower in group C than those in group A and group B （P〈0.05）. Conclusion Intervertebral disc injection with medical ozone can reduce the expression of NF-~B in nucleus pulposus cell of the intervertebral disc cell in ca- nine.
