摘  要:目的探讨NF-κβ、bcl-2/bax基因在百草枯中毒鼠所致急性肺损伤中的表达.方法 80只SD雄性大鼠分为对照组(C组30只)、百草枯中毒组(PQ组50只).PQ组一次性灌胃PQ 25 mg/kg染毒,C组予等体积生理盐水灌胃.观察各组大鼠在中毒后6 h、12 h、1 d、3 d、5 d肺组织病理改变,采用反转录-聚合酶链反应(RT-PCR)方法检测肺组织bcl-2和bax m RNA的表达.采用ELISA方法检测肺泡灌洗液中PMN的NF-κβp65表达.结果 C组肺组织结构完整,无炎症细胞渗出.PQ组肺组织炎性细胞浸润明显增多,且随时间推移肺损伤加重.PQ组bcl-2m RNA的表达于6 h后最强,至3 d仅有少量表;bax m RNA的表达于1 d达高峰,第5天仍有少量表达;bcl-2/bax比例为促凋亡的bax基因占优势,且随着时间的推移这种优势越来越明显;与相应点C组相比差异有统计学意义(P<0.05);PQ组染毒1d时NF-κβp65即有明显表达,3d时表达最强,5d后明显减弱,与C组比较差异有统计学意义(P<0.01).PQ组NF-κβp65与bcl-2表达呈正相关(r=0.71,P<0.01);NF-κβp65与bax表达呈负相关(r=-0.77,P<0.01).结论 NF-κβ、bcl-2/bax介导的肺细胞炎症反应可能是PQ中毒所致急性肺损伤的机制之一.Objective To explore the expressions of nuclear factor kappa B(NF-κβ), apoptosis gene bcl-2(b-cell leukemia-2) and bax(bcl-xl/bcl-2-associated death promoter) in lung of paraquat poisoning rats.Methods Ninety SD male rats were randomly divided into 2 groups: normal control group(C30 rats), paraquat poisoning group(PQ60rats).The PQ group rats were given paraquat by gastric perfusion(25 mg/kg). The C group rats were given saline by gastric perfusion. Histological changes of lung tissues under light microscope were observed.And the expression of bcl-2and bax m RNA in lung tissues of rats were measured by RT-PCR at different time of 6 h,12 h,1 d,3 d,5 d and 7 d after poisoning. The expression of NF-κB p65 in PMN of BALF was determined by ELISA.Results The degree of lung injury in PQ group was higher than that in C group. The relative expression levels of bcl-2 in PQ(6 h and 3 d) subgroups were significantly higher than the control group(P〈0.05).The relative expression levels of bax in PQ(6 h,12 h,1 d,3 d) subgroups were significantly higher. The content of NF-κB p65 in PQ group was higher than that in C group.The expression of NF-κB was positively correlated with that of bcl-2(r =0.71, P 〈0.01), and negatively correlated with that of bax(r =-0.71, P 〈0.01).Conclusion NF- κb and bcl-2/bax-mediated lung tissue cell apoptosis may be one of the major causes which leading to the lung injury in acute paraquat poisoned rats.
