摘  要:目的观察血脂变化对OLETF大鼠胰岛素受体底物-1(IRS-1)和含2个SH结构的蛋白酪氨酸磷酸酶(SH-PTP2)蛋白表达的影响。方法 14只OLETF大鼠被随机分为降脂治疗组和对照组。采用Western杂交技术检测OLETF大鼠肝脏、脂肪组织内IRS-1及SH-PTP2的蛋白表达水平。结果降脂治疗组血糖及血脂水平较对照组下降(P<0.05),肝脏、脂肪组织内IRS-1蛋白表达较对照组增加(t=14.61,15.76,P均<0.01),而SH-PTP2的蛋白表达较对照组减少(t=-3.07、-28.76,P均<0.01)。结论降脂治疗可增加OLETF大鼠肝脏、脂肪组织内IRS-1蛋白表达,减少其SH-PTP2蛋白表达,并可能因此影响其胰岛素信号传导,改善其靶组织对胰岛素的敏感性。Objective To investigate the influence of the changes in lipid on the protein expression of insulin receptor substrate 1 (IRS-1) and protein tyrosine phosphatase with two src-homology 2 (SH- PTP2) in OLETF rats. Methods A total of 14 OLETF rats were randomly divided into lipid-lowering therapy group and control group. The protein expression levels of IRS-1 and SH-PTP2 in liver and adipose tissue were measured by Western blot method. Results The levels of blood glucose and lipid were significantly lower in lipid-lowering therapy group than in control group (P〈0.05). After lipid-lowering therapy, the protein expression of IRS-1 increased significantly in liver and adipose tissue of OLETF rats (t= 14. 61, P= 0.00; t= 15.76, P= 0.00, respectively), but the protein expression of SH-PTP2 decreased (t=-3.07, P=0.02; t =-28. 76, P = 0. 00, respectively). Conclusion Lipid-lowering therapy increases the protein expression of IRS-1 and reduces the expression of SH-PTP2 in liver and adipose tissue, and might therefore affect the insulin signal transduetion and improve the insulin sensitivity of target tissues in type 2 diabetic rats.
