HPLC法测定蛇伤Ⅱ号合剂中大黄酚的含量

摘　　要：目的建立HPLC检测法测定蛇伤Ⅱ号合剂中大黄酚的含量。方法采用C18柱（4.6mm×250mm,5μm）;流动相为甲醇-0.1%磷酸溶液（72：28）,流速为1.0m L·min^-1;检测波长254nm。结果大黄酚在8.580~171.600ng的范围内呈良好的线性关系（r=1.0000）,平均回收率为96.71±0.96（RSD为0.99%）。结论该法具有良好的精密度和重现性,结果准确可靠,可用于蛇伤Ⅱ号合剂中大黄酚的含量测定。OBJECTIVE To establish a method for determination of Chrysophanol in Sheshang Ⅱ mixture by HPLC. METHODS Welch Ultimate XB-C18column（ 4. 6mm × 250 mm,5μm） was used with mobile phase of methanol-0. 1% phosphoric acid solution（ 72∶ 28）. The flow rate was 1. 0m L·min^- 1,and The detection wavelength was254 nm. RESULTS The linear relationship of Chrysophanol was good in the range of 8. 580 ~ 171. 600 ng. The average recovery was 96. 71 ± 0. 96（ RSD0. 99%）. CONCLUSION The method is accuracy and reliable and has good precision and reappearance,which can be used for the measurement of Chrysophilite in Sheshang Ⅱmixture.
