摘 要:【目的】观察胃痞消对脾虚证胃癌前病变(PLGC)大鼠血浆肿瘤坏死因子-α(TNF-α)和白细胞介素-4(IL-4)表达的影响。【方法】将SD大鼠分为6组:正常对照组,模型组,维酶素组(剂量为0.2 g·kg-1·d-1),胃痞消高、中、低剂量组(剂量分别为15、7.5、3.75 g·kg-1·d-1)。采用N-甲基-N’-硝基-N-亚硝基胍(MNNG)溶液(200μg/m L)自由饮用、小承气汤(2 m L/只,生药1 g/m L)灌胃法及饥饱失常等多因素造模18周复制脾虚证胃癌前病变模型,观察各组大鼠的组织病理学及行为学变化,检测血浆TNF-α及IL-4含量。【结果】模型组血浆TNF-α、IL-4含量较正常组显著降低(P<0.05);维酶素组、胃痞消高剂量组、胃痞消低剂量组血浆TNF-α含量较模型组显著升高(P<0.05),模型组与胃痞消中剂量组比较差异无统计学意义(P>0.05);维酶素组、胃痞消高剂量组、胃痞消中剂量组、胃痞消低剂量组血浆IL-4含量与模型组比较均显著升高(P<0.05)。【结论】胃痞消改善脾虚证胃癌前病变大鼠组织学变化的机制可能通过调节血浆TNF-α、IL-4含量变化及胃黏膜间质淋巴细胞及单核细胞的动员而实现。Objective To observe the effect of Weipixiao on the expression of tumor necrosis factor alpha(TNF-α)and interleukin-4(IL-4) in the plasma of rats with gastric precancerous lesions(GPL). Methods SD rats were randomized into six groups, namely normal group, model group, Vatacoenayme group( 0.2 g·kg^-1·d^-1), and high-, middle-and low-dose Weipixiao groups(15, 7.5, 3.75 g·kg^-1-1respedively). The rats had spontaneous intake of N-methyl-N'-nitro-nitrosoguanidine( MNNG) solution(200 μg/mL) combined with irregular diet and gastric gavage of Xiaochengqi Decoction(2 mL for each rat, 1 g/mL) for 18 weeks to induce GPL. The changes of rat behaviors and gastric histological feature were observed, and plasma TNF-α and IL-4 contents were detected. Results The plasma contents of TNF-α and IL-4 in the model group were decreased compared with those in the normal group(P〈0.05). The plasma contents of TNF-α in Vatacoenayme group and high-and lowdose Weipixiao groups were increased compared with those of the model group( P〈0.05), while there was no difference between the model group and middle-dose Weipixiao group(P〉0.05). The plasma content of IL-4 in Vatacoenayme group and high-, middle-and low-dose Weipixiao groups was increased compared with that of the model group(P〈0.05). Conclusion Weipixiao has effects on regulating the expression of TNF-α and IL-4 in the plasma and on activating the interstitial lymphocytes and monocytes, which may be the mechanism for improving the gastric histology of GPL rats with spleen deficiency.
