葛根黄酮自微乳给药体系的制备及质量评价

摘  要:考察葛根黄酮在油相、乳化剂及助乳化剂中的溶解度,采用伪三元相图法优选葛根黄酮自微乳最佳处方。葛根黄酮在油相Maisine 35-1、乳化剂Cremorphor RH-40与助乳化剂1,2-丙二醇中溶解度较大。乳化剂Cremorphor RH-40与助乳化剂1,2-丙二醇质量比3∶2混合时自微乳区最大;油相Maisine 35-1与油相Crodamol GTCC作为混合油相,可增大自微乳区,且二者质量比1∶1时自微乳区最大。葛根黄酮自微乳优化处方各组分的质量比为油相Crodamol GTCC∶油相Maisine 35-1∶乳化剂Cremophor RH-40∶助乳化剂1,2-丙二醇=5∶5∶9∶6,载药量17.65%,平均粒径19.23nm。The solubility of puerarinflavonoids(PF) in various ingredients was investigated, pseudo- ternary phase diagram was employed to op-timize the formula of PF self- mieroemulsifying drug delivery system(PF- SMEDDS). The results showed that PF had a better solubility in theoil of Maisine 35 - 1, the emulsifier of Cremorphor RH - 40 and eo - emulsifier of 1,2 - propanediol. The maximum self- microemulsion regionwas built when Cremorphor RI-I- 40 and 1,2 - propanediol were mixed at mass ratio of 3:2. The mixture of Maisine 35 - 1 and CrodamolGTCC showed an increasing effect on the self- mieroemulsion region and the largest area of self- microemulsion was obtained at their blendingmass ratio of 1 : 1. The formulation of PF- SMEDDS consisted of Crodamol GTCC, Maisine 35 - 1, Cremophor RH - 40, and 1,2 - Propanediolat mass ratio of 5:5:9:6 with 17.65% of drug loading and 19.23um of mean droplet size.
