摘 要:目的探讨黄芪多糖对机体免疫功能的影响,从而为临床应用和相关产品开发提供理论依据。方法每批40只小鼠,随机分为黄芪多糖组1(低剂量),黄芪多糖组2(中等剂量)和黄芪多糖组3(高剂量)以及正常对照组,每组10只。黄芪多糖组每天灌服0.2 ml浓度为5、10、20 mg/ml的黄芪多糖溶液,正常对照组每天灌服0.2 ml0.9%氯化钠溶液,10 d后处死,取样测定小鼠各项免疫指标。固有免疫:检测小鼠巨噬细胞吞噬功能;细胞免疫:小鼠后足迟发型超敏反应强度;体液免疫:血凝法测定小鼠血清中溶血素含量;免疫器官:对脾脏和胸腺称重计算脾脏指数和胸腺指数。结果黄芪多糖3组小鼠巨噬细胞吞噬率和吞噬指数明显高于正常对照组(P<0.01);黄芪多糖3组小鼠左右后足重量差与正常对照组小鼠相比,有明显提高(P<0.01);黄芪多糖3组小鼠抗体积数高于正常对照组(P<0.05);黄芪多糖3组小鼠脾脏指数和胸腺指数均较正常对照组小鼠有明显提高(P<0.01)。结论黄芪多糖可以显著提高机体的固有免疫功能、细胞免疫功能和体液免疫功能。Objective To investigate the effects of Astragalin polysaccharides on immune function of mice in order to provide theoretical basis for clinical application .Methods Forty mice were randomly divided into four groups:low-dose Astragalin polysaccharide group ,median-dose Astragalin polysaccharide group ,high-dose Astragalin polysaccharide group and normal control (NC) group,with 10 mice in each group.The mice in Astragalin polysaccharide groups were given Astragalin polysaccharides 0.2ml by gavage (5mg/ml,10mg/ml,20mg/ml,respectively) per day,however,the mice in NC group were given 0.9%sodium chloride solution 0.2ml by gavage per day .After10 days, the mice were sacrificed without pain and the specimens were collected from them .The immune function parameters of the mice were examined as follows: ( 1 ) Innate immune:macrophage phagocytosis was detected; ( 2 ) Cellular immune: delayed hypersensitivity intensity was tested; ( 3 ) Humoral immune:hemolysin content in serum was examined by hemagglutination test;(4)Immune organ:spleen and thymus indexes were calculated by weighting .Results The phagocyte rate and phagocyte indexes in the three Astragalin polysaccharide groups were significantly higher than those in NC group ( P 〈0.1 ) .As compared with that in NC group ,the weight difference between left and right postpedes in Astragalin polysaccharide groups was significantly improved ( P 〈0.01).The antibody titre in Astragalin polysaccharide groups was significantly higher than that in NC group ( P 〈0.05).As compared with those in NC group , the spleen and thymus indexes in Astragalin polysaccharide groups were significantly increased ( P 〈0.01).Conclusion Astragalin polysaccharides can obviously enhance immune function of organism including innate immune ,humoural immune and cellular immune .
