摘  要:为了探讨葡萄内酯对D-半乳糖致衰老小鼠学习记忆改善作用的影响,为葡萄内酯在抗老年痴呆方面提供理论依据,试验建立D-半乳糖致亚急性衰老小鼠模型,以Morris水迷宫试验进行行为学测试,观察各组小鼠的学习记忆能力,测定各组小鼠脑组织超氧化物歧化酶(SOD)、丙二醛(MDA)、乙酰胆碱酯酶(Ach E)及谷氨酸(Glu)含量的变化。结果表明:葡萄内酯各剂量组逃避潜伏期的时间均能显著缩短,空间探索能力也有明显改善的趋势;且能显著提高脑组织SOD的活性,抑制Ach E的活性,降低MDA的含量。说明葡萄内酯能够改善D-半乳糖致衰老模型小鼠的学习记忆能力和提高脑组织的抗氧化能力,其作用机制可能与抑制脑组织Ach E活性有关。To investigate the effect of auraptene on the improvement of learning and memory in the aging mouse model induced by D- galactose,and to provide theoretical basis for auraptene in terms of anti- senile dementia. The subacute aging mouse model induced by D- galactose was established,and the Morris water maze test was carried out for behavioral test. The ability of learning and memory of the mice in each group were observed,and the changes in the contents of superoxide dismutase( SOD),malondialdehyde( MDA),acetylcholinesterase( Ach E) and glutamic acid( Glu) in the brain tissues of mice were determined. The result showed that the escape latency time of the mice in each dose group with auraptene was significantly shortened,and there was also obvious improving trend in the spatial probe capability of the mice; and it could significantly improve the SOD activity of brain tissue,and inhibit Ach E activity,and decrease the content of MDA. The results indicate that auraptene can improve the ability of learning and memory in the aging mouse model induced by D- galactose,and increase the antioxidant capacity of brain tissue,its mechanism of action may be related to the inhibition of Ach E activity in brain tissue.
