含有性连锁矮小型基因DW和/或dw基因溧阳鸡的检测方法研究

摘　　要：为通过分子生物学手段早期检测矮小型溧阳鸡,通过设计特异性引物,以含有DW和/或dw基因的鸡血液基因组DNA为模板进行PCR扩增,通过琼脂糖凝胶电泳技术进行鉴定。结果显示:只含有DW基因鸡仅扩增出417 bp单一条带;只含有dw基因鸡仅扩增出217 bp单一条带;含有DW和dw基因鸡扩增出417 bp和217 bp两个条带。研究表明,该分子生物学方法能够准确鉴定出含有DW和/或dw基因的溧阳鸡,用于矮小型溧阳鸡的早期选择。In order to detected the dwarf-type Liyang chicken by molecular biologic method,two pairs of specific primers were designed and blood genomic DNA of chicken with DW and/or dw gene was used as template to amplify by PCR. The products were detected by agarose gel electrphoresis. The results showed that chickens only with DW gene had a single band of 417 bp,chickens only with dw gene had a single band of 217 bp,chickens with DW and dw gene had the two bands. This molecular biologic method could identify chickens with DW and/or dw gene. The results provided theoretical foundation for early selection of dwarf type chicken.
