摘  要:目的观察当归芍药散对肝硬化腹水大鼠血浆血管紧张素Ⅱ(angiotensin-Ⅱ,AngⅡ)和内皮素-1(endothelin-1,ET-1)水平的影响。方法将雄性SD大鼠随机分为正常组,模型组,甘利欣组,当归芍药散高、中、低剂量组。采用苯巴比妥灌胃联合四氯化碳腹腔注射复制肝硬化腹水模型。采用酶联免疫吸附试验检测血浆AngⅡ、ET-1水平。结果与正常组比较,模型组大鼠凝血酶原时间显著延长(P<0.05),血浆AngⅡ和ET-1水平显著升高(P<0.05)。与模型组比较,甘利欣组和当归芍药散各剂量组凝血酶原时间均显著缩短(P<0.05),血浆AngⅡ和ET-1水平显著降低(P<0.05)。当归芍药散各剂量组及甘利欣组之间凝血酶原时间及血浆AngⅡ、ET-1水平相比较,差异均无统计学意义(P>0.05)。结论当归芍药散治疗肝硬化腹水的机制与降低血浆AngⅡ、ET-1水平有关。Objective To investigate the effects of Danggui Shaoyao Powder(DSP)on the plasma levels of angiotensin Ⅱ(Ang Ⅱ)and endothelin-1(ET-1)in rats with cirrhotic ascites.Methods Male Sprague-Dawley rats were randomly assigned to normal group,model group,Ganlixin group,and DSP groups which were further divided into high-dose,middle-dose,and low-dose groups.Intragastric administration of phenobarbital and intraperitoneal injection of carbon tetrachloride were adopted to establish a rat model of cirrhotic ascites.Prothrombin time(PT)was measured by a blood coagulation analyzer.The plasma levels of Ang Ⅱ and ET-1 were measured by enzyme-linked immunosorbent assay.Results Compared with the normal group,the model group had significant increases in PT and AngⅡ and ET-1levels(P〈0.05).Compared with the model group,the Ganlixin and DSP groups had significant decreases in PT and Ang Ⅱ and ET-1levels(P〈0.05).PT and AngⅡ and ET-1levels showed no significant differences between the DSP and Ganlixin groups(P〉0.05).Conclusion DSP could reduce plasma levels of Ang Ⅱand ET-1,which may be related to its action mechanism in the treatment of cirrhotic ascites.
