摘  要:目的研究微小核糖核酸-21(miR-21)、miR-31和叉头样转录因子P3(FOXP3)在不明原因复发性自然流产(URSA)患者蜕膜中的表达情况,探讨miR-21、miR-31与FOXP3之间的相关性。方法运用实时荧光定量PCR和蛋白免疫印迹法测定miR-21、miR-31及FOXP3在URSA患者及正常对照组蜕膜组织中的表达情况,并分析miR-21、miR-31的表达与FOXP3表达的相关性。结果FOXP3、miR-21在URSA组中的表达均明显低于正常对照组(P均<0.05),而miR-31在URSA组中的表达高于对照组(P>0.05)。蜕膜组织中miR-21,miR-31的表达与FOXP3的表达相关(P均<0.05),其中miR-21与FOXP3表达呈正相关,而miR-31与FOXP3表达呈负相关。结论蜕膜组织中miR-21和miR-31的表达异常,可能通过下调FOXP3基因的表达而引起母体免疫耐受机制紊乱,从而导致URSA的发生。Objective To investigate the expression of microRNA-21( miR-21),miR-31 and forkhead box P3( FOXP3) and analyze the correlation between miR-21 and miR-31,and FOXP3 in patients diagnosed with unexplained recurrent spontaneous abortion( URSA). Methods The expression levels of miR-21,miR-31 and FOXP3 in the decidua of URSA patients and healthy controls were detected by real-time q PCR and western blot. The correlation between miR-21 / miR-31 and FOXP3 expression was analyzed. Results The expression levels of FOXP3 and miR-21 in the URSA patients were significantly lower than those in the control group( both P 0. 05),whereas the expression of miR-31 in the URSA group was significantly higher than that in the control group. The expression of miR-21 in the decidua tissue was positively correlated with the expression of FOXP3,whereas the expression of miR-31 was negatively associated with FOXP3 expression( both P 0. 05). Conclusion Abnormal expression of miR-21 and miR-31 in the decidua probably causes disorder of maternal immune tolerance mechanism by down-regulating the expression of FOXP3,thereby leading to the incidence of URSA.
