摘  要:目的:观察角膜上皮细胞在细菌脂多糖作用后不同时间Toll样受体(TLR)2、4表达的动态变化,初步明确TLR在角膜细菌感染和保持免疫稳态中的作用。方法:培养永生化人角膜上皮细胞系,分别用10ng/ml和1μg/ml的细菌脂多糖(LPS)刺激,作用2h、4h、8h后检测TLR2、4的表达,4h、8h、12h后检测细胞分泌IL-6能力。结果:不同浓度LPS刺激后,细胞TLR2、4的mRNA表达水平在不同时点均明显增高,但随着刺激时间延长,细胞表面蛋白表达和IL-6的分泌水平则呈现先升后降的趋势。结论:人角膜上皮细胞(HCE)能够表达功能性的TLR2、4,并且在接受刺激后,TLR蛋白表达和炎症因子分泌具有负反馈抑制。Objective:To evaluate the dynamic changes in expression of Toll like receptor on cornea epithelial cells induced by lipopolysaccharide and illustrate the effect of TLR in the bacterial infection and Immune homeostasis.Methods:Immortalized human corneal epithelial cell was treated by 10ng/ml and 1μg/ml lipopolysaccharide.The expression of TLR2 and 4were detected with qRT-PCR from mRNA level and FACS from protein level,at 2,4,and 8hours after stimulation.The IL-6level was examined by ELISA at 4,8,and 12 hours after stimulation.Results:After stimulation with LPS,cells elicited an increase in the content of TLR2 and 4specific mRNA at each time point.The cell surface expression of TLR and IL-6secretion increased at early stage but decreased at late stage of LPS exposure.Conclusion:TLR 2and 4cell surface expression in HCE cell can recognize the stimulation of LPS,and their expression pattern constitutes a negative-feedback loop that is important in the bacterial infection and Immune homeostasis environment in the human cornea.
