摘 要:本研究旨在筛查鸭CD6基因CDS区的SNPs位点及其与血清生化指标和肉质的关联性:随机抽取同日出雏、健康无病、同等管理条件下饲喂10周龄的100只樱桃谷鸭、133只三穗鸭和51只兴义鸭,屠宰采血,测定血清生化指标和肉质,提取DNA,以PCR-SSCP结合直接测序法筛查CD36基因的SNPs位点,分析其与血清生化指标及肉质的关联性。结果表明:仅在樱桃谷鸭CD36基因外显子9中检测到1个SNP位点(g.476593 T>C),表现为3种基因型MM、MN、NN,其中优势基因型MM、优势等位基因M的频率分别为0.453和0.643,位点杂合性较高(H=0.459),呈中度多态(0.25<PIC<0.5),基因型分布严重偏离Hardy-Weinberg平衡(x^2=65.41),在三穗鸭和兴义鸭中未检测到SNPs;进行关联分析发现,突变基因型NN群体除甘油三酯外所有被测血清生化指标均显著高于野生型MM群体(P<0.05),NN基因型群体肌内脂肪显著低于MM型群体(P<0.05),故推测g.476593 T>C突变可能会影响CD36基因的表达活性,使得脂质分解代谢加强,这对今后选育优质低脂肪鸭品种具有重要意义。本研究首次研究了鸭CD36与血清生化指标和肉质的关联性,对今后家禽CD36基因的研究提供重要的参考依据。In this study, the objective was to analysis the SNPs of duck CD36 gene CDS region and its association with serum biochemical levels and meat quality; 100 Cherry valley ducks, 133 Sansui ducks and 51 Xingyi ducks which were which are 10 weeks old, healthy and in the similar environment were selected as the experimental animals, the blood DNA was extracted, we detected the SNPs loci of CD36 gene and analyzed its association with serum biochemical levels and meat quality by PCR-SSCP and sequencing methods; The result showed that the SNP (g. 476593 T〉C ) loci and 3 genotypes (MM, MN, NN) were detected, the dominant genotype and allele was MM and M , the frequency was 0.453 and 0.643 respectively, the loci showed high heterozygosity (h=0.459), the mutation loci belong to moderate polymorphism (0.25 〈PIC〈0.5), and the genotype distribution deviated from Hardy-Weinberg balance seriously (x^22=65.41) , the SNPs was not detected in Sansui ducks and Xingyi ducks, the result of association indicated that all the serum biochemical levels of the mutation genotype NN groups were significant higher than those of the wild genotype MM groups except for triglyceride (P〈0.05), the intramuscular fat of NN groups was significant lower than that of MM groups, so we predicted it is possible that g. 476593 T〉C mutation enhanced the expressing activity of CD36 gene, and made the catabolism of fatty acid be promoted, and it may have an important function that we breed the low fat ducks in future; Moreover, this is the first time to analysis the polymorphism of duck CD36 gene and its association with serum biochemical levels and meat quality, it could provide insight for the study on the CD36 gene in poultry in the future.
