摘 要:目的:测定金钗石斛超细微粉细胞破壁率及超细粉碎前后石斛碱含量。方法:采用细胞计数法,以薄壁细胞为评价指标,测定金钗石斛微粉的细胞破壁率,并用气相色谱法测定超细粉碎前后细粉与微粉石斛碱的含量。结果:金钗石斛细粉称样量与完整薄壁细胞计数的回归分析结果表明,两者呈现显著相关,金钗石斛经超细粉碎后细胞破壁率为100%。利用气相色谱法测定金钗石斛超细粉碎前后细粉与微粉石斛碱含量分别为0.42%、0.35%。结论:此方法简单快速、准确可靠,可准确地进行金钗石斛微粉细胞破壁率及石斛碱的定量测定。Objective: To determine the rate of cell wall-breaking and content of Dendrobine in Dendrobium nobile fines powder and micro-powder. Methods: The cell counting method was used to determine the cell breakage rate of Dendrobium nobile micro-powder, with parenchyma cells as evaluation index. The content of Dendrobine in Dendrobium nobileby was tested by gas chromatography. The weight of Dendrobium micro-powder sample was analyzed and the number of the complete parenchymal cells was counted. Results: The regression analysis showed that the weight of Dendrobium micro-poweder sample and number of complete parenchyral cells had a significant correlation. The cell wall-breaking rate was 100% after Dendrobium nobile was ultrafine grinded. And the content of fines powder and micro-powder were 0.42% and 0.35% respectively. Conclusion:This method is easy, quick and reliable, and can accurately evaluate the rate of cell wall-breaking and content of Dendrobine of Dendrobium nobile micro-powder.
