摘  要:目的研究长期慢性缺氧对紫绀型先天性心脏病患儿自体骨髓间充质干细胞(Bone Marrow-derived Mesenchymal Stem Cells,BMSCs)生物学行为的影响。方法通过血气分析紫绀组和非紫绀组骨髓的氧浓度,检测骨髓血BMSCs含量,评估细胞增殖能力,以及BMSCs定向诱导为成骨、成脂及成软骨细胞的能力。给予缺氧缺血清处理后评估BMSCs的抗凋亡能力。结果紫绀组患儿的骨髓氧分压较非紫绀显著降低(P<0.01),两组骨髓的BMSCs的含量无统计学差异,但紫绀组BMSCs的增殖能力,成骨、成脂、成软骨分化能力及抗凋亡能力较非紫绀组显著降低(P<0.05)。结论长期慢性缺氧能明显降低骨髓的氧浓度,并显著减弱BMSCs的增殖、多向分化及抗凋亡能力,但对BMSCs的含量无影响。Objective To study the effects of long-term chronic hypoxia in vivo of children with cyanotic congenital heart disease on the cell biological behavior of Bone Marrow-derived Mesenchymal Stem Cells. Methods Oxygen concentration of bone marrow was measured through blood gas analysis. The BMSCs ratio was assayed by Aldefluor staining and the abilities of cell proliferation was estimated by MTT. Then BMSCs was differentiated into osteocytes, adipocytes and chondrocytes to evaluate multilineage potential. Apoptosis in response to hypoxia and serum deprivation was also analyzed through flow cytometry. Results The oxygen partial pressure of cyanotic group was significantly lower than non-cyanotic group(26.33±3.06mmHg VS 51.67±5.5 1mmHg, P〈0.01). The BMSCs ratio of two groups showed no statistical differences, while the abilities of cell proliferation and anti- apoptosis of cyanotic group were significantly impaired as well as the abilities of osteogenesis, adipogenesis and chondrogenesis. Conclusion Bone marrow microenvironment of cyanotic children suffered severe hypoxia during long-term chronic hypoxia in vivo, which impaired the BMSCs abilities of proliferation, mullilineage potential and anti-apoptosis except the BMSCs ratio.
