摘  要:【目的】建立天然植物化妆品中3种动植物源致病菌的快速、敏感、特异的多重PCR诊断检验方法。【方法】设计肺炎克雷伯氏菌(Klebsiella pneumoniae)、铜绿假单胞杆菌(Pseudomonas aeruginosa)和地衣芽孢杆菌(Bacillus licheniformisin)3种菌的特异性PCR引物,通过单一、双重及三重PCR反应检测。【结果】种动植物源致病菌的单一、双重和三重PCR均成功扩增出目的片断,没有非特异性扩增。通过对人工感染膏状染发剂的PCR检测进行灵敏度验证,得出三种菌检出限:Klebsiella pneumoniae为4.5×10 CFU/m L;Pseudomonas aeruginosa为6×102CFU/m L;Bacillus licheniformisin为1×10 CFU/m L。【结论】建立的多重PCR反应系统具有较好的特异性和灵敏性,为天然植物型化妆品中常见致病菌的检测提供了快速、简便、可靠的方法。[ Objective ] The project aims at the establishment of multiple PCR fast, sensitive and specific diagnostic test method for the natural plant cosmetics in 3 kinds of animal and plant sources pathogenic bacteri- um, Klebsiella pneumoniae, Pseudomonas aeruglnosa and Bacillus licheniformis. [ Method ] The specific primers of the PCR 3 kinds of bacteria were designed for detection by single, double and three PCR reactions. [ Re- sult] The results showed that 3 kinds of animal and plant pathogens,bothsingle source and three PCR were successfully amplified fragments with no non - specific amplification. Through the hair coloring detection agent PCR on artificial infection of paste sensitivity test, multiplex PCR reaction system was established in this study which showed good specificity and sensitivity: Klebsiella pneumoniae was 4.5 x 10 CFU/mL; Pseudomonas aeruginosa was 6 × 102 CFU/mL;Bacillus licheniformisin was 1 × 10 CFU/mL. [ Conclusion] Multiplex PCR system established in this study had good specificity and sensitivity,which provided a fast, simple and reliable method for the natural plant -based cosmetics common pathogen detection.
