摘 要:为研究反季节生产期鹅催乳素(PRL)和催乳素受体(PRLR)基因表达情况,选取经反季节调控处于产蛋期(试验组)和未调控处于正常休产期(对照组)母鹅,采集垂体、下丘脑和卵巢组织,应用实时荧光定量PCR技术研究PRL和PRLR基因mRNA在下丘脑-垂体-性腺轴中的表达水平变化。结果表明:PRL和PRLR基因在垂体、下丘脑和卵巢组织均有表达,在垂体中表达量最高;PRL基因在试验组鹅垂体中表达量极显著高于对照组,而PRLR基因在试验组鹅垂体中表达量极显著低于对照组;PRL基因在试验组鹅下丘脑中表达量显著高于对照组,而PRLR基因在两组鹅下丘脑中表达元显著差异;PRL基因在两组鹅卵巢中表达量无显著差异,而PRLR基因在试验组鹅卵巢组织中表达量极显著高于对照组。反季节调控可增加鹅垂体和下丘脑中PRL基因表达,抑制垂体PRLR基因表达,增加卵巢中PRLR基因表达。The objective of this study was to investigate the differences of expression of PRL and PRLR gene respond to goose treated by counter-season production.The pituitary,hypothalamus and ovarian were harvested from geese of experimental group and control group.The differences of PRL and PRLR gene expression in hypothalamus-pituitary-gonadal axis were detected by Real-time PCR.The results showed that PRL and PRLR gene was expressed in all three tissues,with the highest expression in pituitary and the lowest in ovarian.In pituitary,the PRL mRNA expression in experimental group was significantly higher than that of control group(P〈0.01),while the PRLR mRNA expression was significantly lower than that of control group(P〈0.01).In hypothalamus,the PRL mRNA expression in experimental group was significantly higher than that of control group(P〈0.05),while the PRLR mRNA expression had no significant difference(P〉0.05).The PRL gene expression in ovarian had no significant difference between the two groups,while the PRLR mRNA expression was significantly higher than that of control group(P〈0.01).Counter-season production treatment increased the expression of PRL gene in pituitary and hypothalamus and decreased the PRLR gene in pituitary and increased in ovarian.
