化痰行瘀汤抗大鼠肝纤维化作用机制研究

摘　　要：[目的]探究化痰行瘀汤抗大鼠肝纤维化作用的可能机制。[方法]取清洁级健康成年Wistar大鼠60只,随机抽取10只作为正常对照组,剩余50只采用橄榄油溶液皮下注射四氯化碳(CCL4)法制作肝纤维化模型,确认造模成功后按随机数字表法分为5组,即模型对照组、复方鳖甲软肝片组、化痰行瘀汤低剂量组、化痰行瘀汤中剂量组、化痰行瘀汤高剂量组。5组分别以相应药物连续干预6W,6W后比较各组Ⅳ型胶原(C-Ⅳ)、层黏连蛋白(LN)、α-平滑肌肌动蛋白(α-SMA)、骨桥蛋白(OPN)和转化生长因子β1(TGF-β1)蛋白、TGF-β1 m RNA的表达。[结果]与正常对照组比较,干预6W后复方鳖甲软肝片组、化痰行瘀汤中剂量组、化痰行瘀汤高剂量组能明显降低C-Ⅳ蛋白的表达(P<0.05);化痰行瘀汤各剂量组LN蛋白表达明显下降(P<0.05);化痰行瘀汤各剂量组与复方鳖甲软肝片组α-SMA表达都明显下降(P<0.05);复方鳖甲软肝片组、化痰行瘀汤低剂量组、化痰行瘀汤高剂量组OPN蛋白表达明显下降(P<0.05);化痰行瘀汤各剂量组TGF-β1蛋白表达明显减少(P<0.05);化痰行瘀汤高剂量组TGF-β1m RNA表达明显下降(P<0.05)。[结论]化痰行瘀汤能有效改善肝纤维化,且与剂量存在一定相关性。[Objective] To observe the therapeutic effects of Hua tan xing yu（HTXY） decoction on experimental hepatic fibrosis and explore its possible mechanism. [Methods] Totally 60 healthy adult Wistar rats of clean degree were selected and randomly assigned into 6 groups with 10 in each group：normal control group,model control group, Fu fang bie jia ruan gan pill group, HTXY decoction groups of low-dose, middle-dose and high-dose. The hepatic fibrosis model was induced by subcutaneous injection of carbon tetrachloride（CCL4） olive oil solution. After the 6 weeks intervention of drugs,the expressions of the C- Ⅳ, LN, α-SMA, OPN, TGF-β1 protein and TGF-β1 m RNA were detected by immunohistochemistry and RT-PCR assay,respectively. [Results] As compared with normal control group, the C- Ⅳ protein expression was significantly lower in Fu fang bie jia ruan gan pill group,HTXY decoction groups of middle-dose and high-dose（P〈0.05）; the LN protein expression was significantly decreased in HTXY decoction groups of low-dose,middle-dose and high-dose（P0.05）;the α-SMA expression was significantly decreased in Fu fang bie jia ruan gan pill group, HTXY decoction groups of low-dose,middle-dose and high-dose（P〈0.05）;the OPN protein expression was significantly decreased in Fu fang bie jia ruan gan pill group,HTXY decoction groups of low-dose and high-dose（P〈 0.05）; the TGF-β1 protein and m RNA expression were significantly decreased in HTXY decoction groups of low-dose,middle-dose and high-dose（P0.05）.[Conclusions] HTXY decoction can effectively relieve liver fibrosis.
