摘 要:[目的]观察人参皂苷对桥本甲状腺炎大鼠甲状腺Foxp3、T-bet、GATA-3 m RNA表达的影响。[方法]40只Wistar大鼠随机分为五组,即正常组、桥本甲状腺炎模型组、人参皂苷低、中、高剂量组,治疗组人参皂苷灌胃,正常及模型组以蒸馏水灌胃。用酶联免疫法检测甲状腺自身抗体水平,HE染色观察甲状腺组织病理改变,RT-PCR检测甲状腺组织Foxp3(Forkhead/winged helix transcription factor,叉状头/翅膀螺旋转录因子)、T-bet(T-box expressed in T cells,转录因子T-bet)、GATA-3(GATA binding protein 3,转录因子GATA-3)的表达。[结果]与正常组相比,模型组的TGAb(Thyroglobulin,甲状腺球蛋白抗体)及TPOAb(Thyroid peroxidase antibody,甲状腺过氧化物酶抗体)水平显著增加(P<0.05),T-bet m RNA表达水平也显著增加(P<0.05),Fox P3、GATA-3 m RNA表达显著下调(P<0.05);与模型组相比,人参皂苷治疗组的TGAb及TPOAb均下降,同时T-bet m RNA表达下降,其中低剂量与中剂量组有统计学意义(P<0.05),而人参皂苷治疗各组的GATA-3、Foxp3的表达水平均有所上升,其中低剂量与中剂量组有统计学意义(P<0.05)。[结论]Fox P3、GATA-3 m RNA表达下调,T-bet m RNA表达上调可能是桥本甲状腺炎发病的重要原因之一。人参皂苷可能通过上调GATA-3、Foxp3的表达,下调T-bet的表达,抑制Th1细胞功能的过度亢进,调节免疫的途径来保护甲状腺组织。[Objective]To investigate the effects of ginsenoside on the expression of Foxp3, T-Bet and GATA-3 in Hashimoto's thyroiditis rats. [Methods]40 Wistar rats were divided into five groups randomly, they were normal group,the model control group,low dose group of ginsenosides, middle dose group and high dose group of ginsenosides. Thyroid autoantibodies detected by enzyme-linked immunosorbent assay.Pathological changes in rat thyroid tissue by HE staining. Foxp3(Forkhead/winged helix transcription factor, forked head/wings helix transcription factor), T-bet(T-box expressed in T cells,the transcription factor T-bet), GATA-3(GATA binding protein 3, transcription factor GATA-3) expression in rat thyroid tissue were detected by RTPCR. [Results]Compared with the normal group, TGAb(Thyroglobulin, thyroglobulin antibodies) and TPOAb(Thyroid peroxidase antibody, thyroid peroxidase antibody) in model group were increased, as well as T-bet m RNA level( P〈0.05),and GATA-3, Foxp3 m RNA level were deceased(P〈0.05).Compared with model group, ginsenoside treatment declined the TGAb and TPOAb level, while the m RNA expression of T-bet also decreased,especially in which low and middle dose groups(P〈0.05), and GATA-3, Foxp3 expression levels in all the ginsenoside treatment groups rose, especially in low-dose and middle-dose groups were statistically significant( P〈0.05). [Conclusion]Downregulation of Fox P3, GATA-3 and up-regulation of T-bet may be the important reasons for the onset of Hashimoto's thyroiditis. Ginsenoside may take effect in Hashimoto's thyroiditis rats by reducing the expression of T-bet and increasing the expression of GATA-3 and Foxp3, and through suppression Th1 cell hyperfunction.
