隔药饼灸对动脉粥样硬化兔主动脉内皮细胞NF-κ BmRNA及MMP-2mRNA、MMP-9mRNA的影响

摘　　要：目的观察隔药饼灸对动脉粥样硬化(artherosclerosis,AS)兔主动脉内皮细胞核因子-κB(nuclear factor-kappa B,NF-κ B)转录RNA及基质金属蛋白酶-2、9(matrix metalloproteinases,MMP-2、9)转录RNA的影响,探讨隔药饼灸对动脉粥样硬化(AS)形成的干预作用。方法将75只新西兰大耳白兔随机分为5组,即:空白组(空白对照组)15只;模型组(AS模型组)15只;直接灸组(AS模型+艾炷直接灸)15只;隔药饼灸组(AS模型+隔药饼灸)15只;西药组(AS模型+阿托伐他汀)15只。除空白组外,其余4组通过高脂饲料喂养及免疫损伤法建立兔AS模型。分别运用RT-PCR技术、原位杂交法检测兔主动脉内皮细胞NF-κ BmRNA、MMP-2、9mRNA的表达。结果直接灸、隔药饼灸和西药均能抑制动脉粥样硬化兔主动脉内皮细胞NF-κ BmRNA、MMP-2、9mRNA的表达(P<0.01或P<0.05);隔药饼灸和西药抑制作用明显优于直接灸(P<0.01或P<0.05)。结论隔药饼灸能够起到抗AS的作用,可能是通过抑制NF-κB的核结合活性,进而降低MMP-2、9的表达完成的。Objective To observe the influence of moxibustion with herbal cake on nuclear factor-kappa B（ NF-κB） and matrix metalloproteinases-2,9（ MMP-2,9） in aortic endothelial cells of atherosclerotic rabbits and investigate whether moxibustion with herbal cake could delay artherosclerosis formation. Methods 75 New Zealand white rabbits were randomly divided into 5 groups： Blank group,model group, direct moxibustion group, moxibustion with herbal cake group and western medicine group, 15 rabbits in each group.Expect blank group, rabbit as model was developed by high fat diet and immune injury method. NF-κBm RNA in aortic endothelial cells were measured by reverse transcription polymerase chain reaction（RT-PCR） and the m RNA expression of MMP-2,9 were detected by the method of in situ hybridization. Results Direct moxibustion, moxibustion with herbal cake and western medicine can all decrease contents of NF-κB 、 MMP-2,9 in Aortic Endothelial Cells of Atherosclerotic Rabbits（ P 0.01 or P 0.05）; while the effects of moxibustion with herbal cake and western medicine were more significant than direct moxibustione（ P 0.01 or P 0.05）. Conclusion Moxibustion with herbal cake can decrease the expression of MMP-2,9 by inhibiting the activation of NF-κB to delay artherosclerosis formation.
