摘  要:目的:建立测定清肠化湿颗粒中盐酸小檗碱含量的方法。方法:采用HPLC法,色谱柱:Hedera C18柱(250×4.6 mm,5μm),以乙腈-0.05 mol·L-1磷酸二氢钾溶液(50∶50,每100 m L中加十二烷基硫酸钠0.4g,再以磷酸调节p H值为4.0)为流动相,流速为1.0 m L·min-1,检测波长为345 nm,柱温为30℃。结果:盐酸小檗碱的检测浓度在3.8125-122.0μg·m L-1范围内与峰面积积分值呈良好的线性关系(r=1.0000),平均加样回收率为99.85%,RSD%=1.83%(n=9)。结论:HPLC法简便易行,结果准确,重现性好,可用于测定清肠化湿颗粒中盐酸小檗碱含量。Objective: To establish a method for the determination of berberine hydrochloride in Qingchanghuashi Granule(清肠化湿颗粒) by HPLC. Methods: The Hedera C18column(250 mm×4.6 mm,5 μm)was used, acetonitrile-0.05 mol·L-1 potassium dihydrogen phosphate(50∶50)(0.4 g Sodium dodecyl sulfate add in each 100 m L solution, with phosphoric acid to adjust p H4.0)as the mobile phase. The flow rate was 1.0 m L·min-1,and the detection wavelength was 345 nm. Results: The linear range of Berberine hydrochloride was 3.8125-122.0 μg·m L-1(r=1.0000),the average recovery rate was 99.85%,and RSD was 1.83%(n=9).Conclusion: The method is simple, rapid, accurate and reliable. It can be used for the content determination of Berberine hy-drochloride in Qingchanghuashi Granule.
