健脾益肺口服液对慢性阻塞性肺疾病肺脾气虚证大鼠核因子-κB的影响

摘　　要：目的:观察健脾益肺口服液对慢性阻塞性肺疾病(chronic obstructive pulmonary disease,COPD)"肺脾气虚证"大鼠肺组织核因子-κB活性的影响,探讨其是否通过核因子-κB途径抑制COPD的气道炎症及调节其免疫功能。方法:将65只SD成年雄性大鼠随机分为6组,分别为空白组,健脾益肺口服液低剂量组、中剂量组、高剂量组,模型组,地塞米松组;用香烟烟熏加脂多糖复制"肺气虚证"模型;接着用大黄液灌胃,复制"脾气虚证";造模时间共为28 d,在第14天开始给药,造模后取大鼠肺组织,用试剂盒提取组织蛋白质,利用Western Blot方法,测定大鼠肺组织I-κB及核因子-κB的表达水平。结果:1模型组大鼠核因子-κB显著高于正常组(P<0.01),I-κB显著低于正常组(P<0.01)。2健脾益肺口服液各剂量组及地塞米松组大鼠核因子-κB表达明显下降,与模型组比较,差异均有统计学意义(P<0.05或P<0.01);健脾益肺口服液高剂量组、中剂量组及地塞米松组I-κB显著升高,与模型组比较,差异有统计学意义(P<0.05或P<0.01);随剂量的增加,疗效亦增加,但健脾益肺口服液各组与地塞米松组之间差异无统计学意义(P>0.05)。结论:健脾益肺口服液可增强I-κB活性,抑制COPD"肺脾气虚证"大鼠核因子-κB,从而推断其可能通过抑制核因子-κB改善COPD免疫功能,抑制其气道炎症。Objective： To observe the influence of jianpi yifei liquid on NF-κB of COPD with lung-spleen qi deficiency syndrome in rats,and to explore if its function of inhibiting airway inflammation and adjusting immune function is through the NF-κB pathway in COPD.Methods： 65 SD rats were randomly divided into six groups,blank control group,jianpi yifei liquid groups in small,middle and large dosage,model group and dexamethasone group. Rats were treated with smoke and lipopolysaccharide（ LPS） for the attern of lung-qi deficiency. Then from gavaging rats with rhubarb liquid,the model of spleen-qi deficiency was derived. Model-making lasted for 28 days.Rats were administrated for 14 days. After model-making,lung tissue was taken,tissue proteins were extracted by the kit,and then expressions of I-κB and NF-κB were determined with western blotting method. Results： 1. The expression rate of NF-κB of model group was significantly higher than those of normal control group（ P〈0. 01）,the expression rate of I-κB of model group was significantly lower than those of normal control group（ P〈0. 01）; 2. The expression rate of NF-κB in jianpi yifei liquid every-dosage group and dexamethasone group were notably increased,lower than the normal control group（ P〈0. 05,P 0. 01）; Expression rate of I-κB in jianpi yifei liquid large dose,middle dose and dexamethasone group were notably increased,higher than the normal control group（ P〈0. 05,P 0. 01）; With the increase of the dose,the curative effect was also increased,but the treatment groups and dexamethasone group showedthat there was no statistically significant difference between four groups（ P〈0. 05）. Conclusion： Jianpi Yifei Liquid can increase I-κB activity and reduce expression of NF-κB during COPD with lung-spleen Qi deficiency syndrome in rats,and we could infer that Jianpi Yifei Liquid might inhibit airway inflammation and adjust immune function through the NF-κB signaling pathway in COPD.
